摘  要:本研究用12对引物组合对杂交水稻生产中广泛应用的优良杂交组合及其亲本(共15个材料)进行了AFLP分析。通过优选,筛选出 3对引物,即 E-ACT/M·CAC, E-AAC/M-CAA及 E-AAG/M-CAA,它们是本研究中用来区分所研究的 15个材料所需的最少引物组合。然后根据所选用的16个多态性DNA条带的有无,把每个材料的DNA指纹转换成由1和0组成的指纹数据。在此基础上,开发了一套应用软件。在实际应用中,将样品的扩增结果转换成指纹数据,由计算机辅助进行种子鉴定。AFLP analysis of 15 rice varieties that were widely used in rice production in China was carried out 3 primer-pairs, i. e. E-ACT/M-CAC, E-AAC/M-CAA 及 E-AAG/M-CAA were screened out. They were the minimum number of primer pairs to be used to distinguish the 15 analyzed rice varieties. DNA fingerprints of the 15 varieties were developed based on 16 selected band amplified by the above three primer-pairs. The DNA fingerprint of each variety was converted into a computerized fingerprint constituted of 1 and 0 based on the presence or absence of the DNA band. In the practical seed identification, the AFLP-DNA fingerprint of a sample was converted into the computerized fingerprint, which can be processed by the software SIR designed by us, so the seed identitication can be completed by the aid of a computer.
