摘 要:目的对分别以热熔压敏胶(简称热熔胶)和橡胶为基质的两种关节止痛膏的药物体外透皮特性、药效以及皮肤刺激性进行比较研究。方法采用裸鼠腹部皮肤,用Franz扩散池考察不同剂型中药物的体外透皮特性;以热板实验和扭体实验考察两种剂型对小鼠的镇痛效果;以小鼠耳廓肿胀和大鼠足肿胀实验考察两种剂型的抗炎效果;以豚鼠皮肤损伤实验考察两种剂型对皮肤的刺激性。结果以热熔胶为基质的关节止痛膏有更好的药物体外透皮特性和更强的镇痛抗炎效果,对皮肤的刺激性也小于橡胶膏剂型。结论热熔胶是一种制备中药贴膏的理想基质,具有透皮快、疗效好、皮肤刺激性小的特点。Objective To compare the differences in transdermal permeability, pharmacodynamics, and skin irritation of Guanjie Zhitong Patches based on two different formulations taking hot-melt pressure sensitive adhesive and rubber as matrixes. Methods In vitro transdermal permeation was performed by means of Franz diffusion cells; The analgesic effect was evaluated by HAc-induced writhing response in mice and hot-plate test, while the anti-inflammatory action was assessed by xylem-induced ear edema in mice and albumin-induced paw edema in rats. The skin irritation of the two matrixes was investigated using guinea pig as model animal. Results Patches based on hot-melt pressure sensitive adhesives showed better transdermal permeability, analgesic, and anti-inflammatory effects but less skin irritation than the rubber type patches. Conclusion Hot-melt pressure sensitive adhesive is a kind of appropriate material to compose the patch matrix for Chinese herbal medicine, which has the good transdermal permeability and efficacy and less skin irritation.
