摘  要:根据绵羊毛囊角蛋白关联蛋白(KAP)6.1(登录号为AY483218.1)基因序列设计合成2个特异性引物,以新疆细毛羊为研究对象,从采集的皮肤组织中提取RNA进行RT-PCR反应,扩增出长度为320 bp的特异性片段,回收目的片段并连接至pMD18-T载体后转化大肠杆菌DH5α感受态细胞,检测阳性克隆、测序并进行分析。结果表明:试验成功克隆了新疆细毛羊KAP6.1基因,得到的长度为320 bp序列中包含252 bp的编码区序列。新疆细毛羊与绵羊亲缘关系最近,同源性为99.2%,与山羊同源性为94.8%,有13处核苷酸差异与人和兔的同源性分别为80.8%、75.1%。Two specific primers were designed according to the gene sequences of keratin - associated protein (KAP) 6.1 of sheep hair follicle (accession number AY483218.1 ). Xinjiang fine -wool sheep were selected as the research object to extract the RNA from the collected skin tissue, and the amplification of 320 - bp specific fragment was performed by using RT - PCR. The recovered fragment was connected with the pMDI8 -T vector, aild then transformed into DI-ISot to test the positive clones for sequencing and analyzing. The results showed that the KAP6. 1 gene was successfully cloned, and a 320 - bp sequence was obtained, which contained a 252 - bp coding region sequence. The genetic relationship of Xinjiang fine - wool sheep was closest to sheep, its sequence homology was 99.2%. The sequence homology of Xinjiang fine - wo01 sheep with goat was 94.8% ,and there were 13 nucleotide differences. The sequence homologies of Xinjiang fine - wool sheep with human and rabbit were 80.8% and 75.1%, respectively.
