赛拉菌素溶液对靶动物犬的安全性研究

摘　　要：为检验赛拉菌素溶液对靶动物德国牧羊犬的安全性,试验将40只德国牧羊犬(公母各20只)随机分为5组,每组8只犬(公母各4只)。1组为赛拉菌素溶液正常剂量组(6 mg/kg),2,3,4组分别为低剂量组(18 mg/kg)、中剂量组(30 mg/kg)、高剂量组(60 mg/kg),5组为空白对照组,分别于给药前和给药后第5,12,30天每只犬的前肢股静脉采集全血4 mL和抗凝血2 mL,进行肝肾功能血液生化指标和血液学指标检测;并在试验30 d后处死犬,剖检取心脏、肝脏、肺脏、肾脏进行病理学检查。结果表明:所有试验犬在给药前后各项血液学指标均在正常值范围内,没有明显变化,差异不显著(P>0.05);给药前后肝肾功能的血液生化指标均在正常范围内,并且给药前后各项指标变化差异不显著(P>0.05);各组试验犬心脏、肝脏、肺脏、肾脏等实质器官病理检测结果无明显异常。说明赛拉菌素制剂在6～60 mg/kg范围内使用时,对机体的肝脏、肾脏未造成明显的毒副作用,提示在试验剂量范围内对犬无明显临床可观察毒性作用。To test the safety of selamectin solution in German Shepherd dog,40 German Shepherd dogs （20 each male and female） were random- ly divided into five groups of 8 dogs each. Group 1 was normal dose group （6 mg/kg BW）, group 2,3 and 4 were low - dose group（ 18 mg/kg BW ）, middle - dose group （ 30 mg/kg BW） and high - dose group （ 60 mg/kg BW）, and group 5 was blank control group. The samples of 4 mL whole blood and 2 mL anticoagulant blood were collected from femoral vein in the foreleg of the each dog before administered and after administered for 5 days, 12 days and 30 days, and then were used to detect blood biochemical and hematological indices for liver and kidney function. At 30 days after the administration, the dogs were sacrificed and dissected their heart, liver, kidney, lung and spleen for pathological examination. The result showed that for all test dogs, the hematological indices were within the normal range before and after administration, without any significant changes （ P 〉 O. 05 ）. The blood biochemical indices for liver and kidney function were within the normal range before and after administration, and their differences were not significant （P 〉 O. 05 ）. The pathological examination showed that there were no obvious abnor- malities in the heart, liver, kidney, lung and spleen of the test dogs in each group. The results indicate that selamectin solution used in the range of 6 to 60 mg/kg, causes no significant toxic side effects on the liver and kidney of the body, suggesting that there is no clinically significant toxic effects in dogs which can be observed within the range of test dose.
