3味山姜属中药不同提取物对胃实寒证大鼠胃黏膜的保护作用  

Protective Effects of Different Extractions from Three Kinds Alpinia of Traditional Chinese Medicines on Stomach Mucous Membrane in Rat Stomach Cold Models

摘　　要：目的:研究山姜属中药高良姜、草豆蔻、红豆蔻对胃实寒证模型大鼠胃黏膜保护作用,揭示它们所含挥发油与温中散寒功效的相关性,以及以水煎剂入药的科学性。方法:将220只大鼠随机分为空白组,模型对照Ⅰ、Ⅱ组,阳性对照组,受试药物(高良姜、草豆蔻、红豆蔻)挥发油高、低剂量组,水煎液高、低剂量组,去挥发油水液高、低剂量组,共22组,每组10只。采用灌服寒凉药(石膏、知母、黄柏、龙胆草)的方法造模,给药剂量为30 g.kg-1,每日2次,连续给药2 d。分别予以高良姜、草豆蔻、红豆蔻挥发油、水煎液、去挥发油水液灌胃,分高(60 g.kg-1)、低(20 g.kg-1)剂量组,每隔12 h给药1次,连续给药3次。阳性对照药选用附子水煎液8 g.kg-1。光镜下观察胃黏膜的病理学改变,采用Mann-whitney秩和检验进行统计分析。结果:高良姜挥发油高、低剂量组,水煎液高剂量组,去挥发油高剂量组,草豆蔻3种提取物各剂量组,红豆蔻3种提取物各剂量组对炎细胞浸润有显著改善作用(P<0.01或P<0.05);高良姜挥发油高剂量组、水煎液高剂量组、去挥发油高剂量组,草豆蔻挥发油高、低剂量组,水煎液高剂量组,去挥发油高、低剂量组,红豆蔻挥发油高、低剂量组,水煎液高、低剂量组,去挥发油高剂量组对腺体减少或破坏有显著改善作用(P<0.01或P<0.05);高良姜挥发油高、低剂量组,水煎液高、低剂量组,草豆蔻3种提取物各剂量组,红豆蔻挥发油高、低剂量组,水煎液高剂量组,去挥发油高剂量组对黏膜充血水肿有显著改善作用(P<0.01或P<0.05)。结论:高良姜、草豆蔻、红豆蔻各给药组对胃实寒证大鼠胃黏膜有不同程度的保护作用,这3味药的主要活性成分是挥发油,水煎液、去挥发油水提液中也含有有效成分,以水煎液入药具有科学依据。Objective：To study the protective effects of Alpiniae Officinarum Rhizome, Alpiniae Katsumadai Semen and Galangae Fructus on stomach mucous membrane in rat stomach cold models, revealing the correlation between the volatile oil in these Chinese medicine and the efficacy of warming middle-jiao to dispel cold, explaining the scientific connotation of medication by boiling water extracts. Method： Two hundred and twenty rats were randomly divided into 22 groups： blank group, model groupⅠand Ⅱ, positive group, high and low dose volatile oil groups, high and low dose boiling water extract groups, high and low dose boiling water extract without volatile oil groups. All rats except blank group were fed cold drugs in the dose of 30 g·kg-1 twice a day for two days. Then they were given different volatile oil, boiling water extract and boiling water extract without volatile oil of the three kinds of Chinese medicines. The pathological changes of the stomach mucous membrane were observed under optical microscope. Result： The high and low dose volatile oil groups and high dose boiling water extract group and high dose boiling water extract without volatile oil group of Lesser Galangal Rhizome, all the groups of Katsumada Galangal Seed and Galanga Galangal Fruit could significantly improve the inflammatory cell infiltration（P〈0.05 or P〈0.01）.
