抵当汤早期干预对糖尿病大鼠视网膜ICAM-1和VCAM-1表达的影响

摘　　要：目的通过链脲佐菌素(STZ)诱导建立糖尿病大鼠模型,探讨不同时间段给予抵当汤干预对糖尿病大鼠视网膜病变形态学影响及其作用机制。方法利用STZ尾静脉注射,配合高脂饲料饮食制备糖尿病大鼠模型,将大鼠分为正常对照组和造模组,造模组又分为模型对照组、抵当汤晚期组、抵当汤中期组、抵当汤早期组、辛伐他汀组、马来酸罗格列酮组。观察抵当汤对糖尿病大鼠体重、血糖、血胰岛素的影响,HE染色后观察视网膜形态学改变,免疫组化染色观察细胞间黏附分子-1(ICAM-1)和血管内皮细胞黏附分子-1(VCAM-1)蛋白的表达。结果实验第13周,与正常对照组比较,造模组空腹血糖明显升高,差异有统计学意义(P<0.01);与模型对照组比较,抵当汤早期组大鼠视网膜HE染色病变相对轻微;与模型对照组比较,抵当汤早期组大鼠视网膜免疫组化染色ICAM-1和VCAM-1表达减少(P<0.01)。结论抵当汤早期干预可延缓糖尿病大鼠视网膜病变的发展。Objective To explore morphological effects and its action mechanism of intervention with Didang decoction at different time periods on retinopathy in diabetic rats through streptozotocin（STZ）-induced diabetic rat model.Methods With caudal vein injection of STZ and high fat diet to make diabetic rat model,all the rats were divided into a normal control group and a model group,and the model group was divided into a model control group,a late stage of Didang decoction group,a middle stage of Didang decoction group,an early stage of Didang decoction group,a simvastatin group and a rosiglitazone maleate group.The effects of Didang decoction on diabetic rats’weight,blood glucose and insulin,as well as,the morphological change in retina after HE staining and expression of intercellular adhesion molecule-1（ICAM-1） and vascular cell adhesion molecule-1（VCAM-1） by immumohistochemical staining were observed.Results At the 13th week of the experiment,compared with the normal control group,fasting blood glucose in the model group was obviously increased with statistical significance（P 0.01）;Compared with the model control group,the rats in the early stage of Didang decoction group had relatively mild retinopathy of HE staining.Compared with the model control group,the expression of ICAM-1 and VCAM-1 of immumohistochemical staining in the rats’retina was decreased in the early stage of Didang decoction group（P 0.01）.Conclusion The early intervention with Didang decoction could delay the development of retinopathy in diabetic rats.
