摘  要:目的探讨促肝细胞生长因子(HGF)对脓毒血症诱导的大鼠多器官损伤的保护作用。方法将雄性SD大鼠分为对照组、假手术组、模型组、HGF治疗组、血红蛋白加氧酶-1(HO-1)抑制剂组及HO-1抑制剂+HGF治疗组。采用盲肠结扎穿孔法(CLP)制大鼠脓毒血症模型。尾静脉注射给药,2次/d;HO-1抑制剂及HO-1抑制剂+HGF治疗组于给予HGF后2 h静脉注射锌原卟啉。术后72 h处死存活大鼠,取血清及主要脏器检测相关指标。结果静脉注射HGF(75、150μg/kg.d)能够降低脓毒血症大鼠死亡率,显著降低血清IL-1β水平(P<0.05);明显改善脓毒血症对大鼠肺、肾和肠的病理改变,增强肺、肾和肠中SOD活性。HO-1抑制剂锌原卟啉可部分拮抗HGF的保护作用。结论 HGF对脓毒血症造成大鼠多器官损伤具有保护作用。Objective To investigate the protective effect of hepatocyte growth promoting factors (HGF) on multiple organ injuries of sepsis in rats. Methods The male SD rats were injected twice a day with either HGF or saline i.v. before and af- ter cecal ligation and puncture operation were done. An HO - 1 inhibitor (Znpp) was injected 2 h after administering HGF. The rat mortality in each group within 72 hours after operation was calculated and the survived rats were killed to get blood and lung, kidney and intestine. Results Compared with sepsis model, HGF (75 μg/kg · d and 150 μg/kg · d) de- creased the mortality dose - dependently in rats. The level of IL - β1 serum that treated by HGF wassignifieantly lower than that in sepsis. The pulmonary, renal and intestinal pathological injuries were more ameliorated in HGF groups. The HO - 1 inhibitor could enhance the injuries and diminished the protective effects of HGF on CLP - induced multiple organ inju- ries. Conclusion HGF could inhibit multiple organ injuries induced by sepsis.
