塞来昔布对体外幽门螺杆菌诱导GES-1转化细胞抑制作用的研究

摘　　要：目的研究选择性环氧合酶(COX)-2抑制剂塞来昔布对幽门螺杆菌(Hp)诱导的人永生性胃上皮细胞株(GES-1)转化细胞是否有抑制增殖、诱导凋亡的作用。方法在体外将幽门螺杆菌悉尼株(SS1)与GES-1细胞共培养建立GES-1细胞转化模型,用四甲基偶氮唑蓝(MTT)法、平板克隆形成实验检测细胞转化情况;再利用MTT法、流式细胞仪检测观察塞来昔布对转化细胞的增殖及凋亡的影响。结果与GES-1对照细胞相比,Hp(SS1)与GES-1细胞共同培养2个月后细胞增殖活性明显增高。塞来昔布作用于GES-1转化细胞,随着药物浓度的升高(50、75、100μmol/L),GES-1转化细胞增殖活性逐渐降低,凋亡率逐渐升高。结论幽门螺杆菌长期作用于胃上皮细胞,可以诱导其转化;塞来昔布可抑制GES-1转化细胞的增殖,并促进其凋亡。Objective To investigate the effect of selective cyclooxygenase-2 （COX-2） inhibitor （celecoxib） on the in- hibited proliferation and induced apoptosis in gastric epithelial cell transformation induced by Helicobacter pylori （H.pylori）. Methods In vitro, the immortalized ＇normal＇ gastric epithelial cell line GES-1 was co-cultured for two months with H. py- loft Sydney strain （SS1） to establish the GES-1 transformation cell model. The cell transformation was measured by 3-（4,5-di- methylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） assay and colony formation assay. The proliferation and apopto- sis of transformed ceils treated by celecoxib were detected by MTF assay and flow cytometry. Results The proliferative activ- ity of GES-1 cells co-cultured with H. pylori （SS1） for two months was significantly higher compared with that of control GES-1 cells. The proliferative activity of GES-1 transformed cells was inhibited with the increased concentration of celecox- ib （50, 75 and 100 μmol/L）, and the apoptotic rate of the GES-1 transformed ceils was gradually increased. Conclusion The transformation of gastric epithelial cells can be induced by H. pylori for a long term. Celecoxib inhibits the proliferation and induce apoptosis of GES-1 transformed cells.
