摘  要:目的通过脑脊液microRNA检测,初步筛选重型颅脑创伤后昏迷患者的相关生物标志物。方法选取颅脑创伤后昏迷(GCS≤8分)患者的脑脊液及正常压力脑积水患者脑脊液标本作为正常对照,利用芯片系统进行microRNA表达谱检测,分析表达出现变化的microRNA类型。结果与对照相比较,颅脑创伤昏迷后患者脑脊液中10种microRNA表达上调P〈0.05,4种microRNA表达下调P〈0.05。结论颅脑创伤后昏迷患者脑脊液中出现表达显著变化的microRNA类型,这些小分子物质的变化对于分析颅脑创伤患者昏迷的机制以及特异性治疗目标提供了进一步分析的研究基础。Objective To screen microRNA related biomarkers of traumatic coma through profile array determination from the cerebral spinal fluid. Methods Patients of severe traumatic brain injury were recruited following critical evaluation and the cerebral spinal fluids were detected for the expression of microRNA( s ) compared with normal pressure. Results Significant differencesin micro RNAexpressions were identified in the cerebral spinal fluid of severely injured patients who were still in the comatose state two weeks after the traumatic insult. 10 microRNAs were found with high expression and 4 microRNAs with decreased expression P 〈 0. 05. Conclusions MicroRNAs detected in the cerebral spinal fluid of severe traumatic coma patients may indicate a unique subtype of biomarker related with unconsciousness as a consequence of brain trauma, not only to the end of understanding the mechanism but the therapeutic target as well.
