摘  要:目的观察转化生长因子β1(transforminggrowth factor beta 1,TGF-β1)和碱性成纤维细胞因子(basic fibroblast growth factor,bFGF)在伴或不伴变应性鼻炎(AR)的鼻息肉组织中的表达情况,探讨它们在鼻息肉发病机制中的可能作用以及它们在鼻息肉中的相关性。方法采用免疫组织化学技术EnVision二步法检测TGF-β1和bFGF在30例不伴AR的鼻息肉组织(非变应性组)、30例伴AR的鼻息肉组织(变应性组)、1 0例鼻中隔矫正术时切除的中鼻甲黏膜组织(正常对照组)中的表达。测定阳性表达的平均光密度(平均光密度与阳性程度成反比),采用SPSS13.0软件进行统计学分析。结果①TGF-β1和bFGF在非变应性组和在变应性组的表达明显高于正常鼻黏膜,主要表达于腺体细胞、上皮细胞和内皮细胞、炎性细胞的细胞浆或细胞核。②对切片的阳性表达测定平均光密度进行统计学分析显示:TGF-β1在非变应性组的表达(151.43±7.67)和在变应性组的表达(147.51±6.97)明显强于在正常对照组(181.79±2.44)的表达,差异具有统计学意义(P<0.05);TGF-β1在变应性组的表达(147.51±6.97)也强于其在非变应性组的表达(151.43±7.67),两者差异具有统计学意义(P<0.05)。③bFGF在非变应性组的表达(147.33±9.86)和在变应性组的表达(145.25±7.66)明显强于在正常对照组(184.74±2.27)的表达,差异具有统计学意义(P<0.05);bFGF在变应性组的表达(145.25±7.66)强于其在非变应性组的表达(147.33±9.86),两者差异不具有统计学意义(P>0.05)。④TGF-β1和bFGF在非变应性组的表达有显著的正相关关系,具有一致性(r=0.368,P<0.05);TGF-β1和bFGF在变应性组的表达有显著的正相关关系,具有一致性(r=0.442,P<0.05)。结论①TGF-β1和bFGF在鼻息肉组织中的表达增高,可能对鼻息肉的形成发展共同起作用;②变应性因素的影响可能加重了TGF-β1和bFGF在鼻息肉中的表达。OBJECTIVE To confirm the expression and distribution of transforming growth factor beta 1 (TGF-β1) and basic fibroblast growth factor (bFGF) in nasal polyps (Group NAR and Group AR) and normal nasal mucosa, to explore their possible role in the development of nasal polyps with or without allergic rhinitis. METHODS TGF-β1 and bFGF were studied in 30 samples of nasal polyp without allergic rhinitis (non-allergic group) , 30 samples of nasal polyp with allergic rhinitis (allergic group) and 10 normal nasal mucosa (control group) with the method of immunohistochemical staining. And the average optical density of positive expression cells were measured (average optical density has a negative correlation to the positive expression) . The SPSS13.0 software was used for the statistical analysis. RESULTS 1. The expression of TGF-β1 and bFGF in non-allergic group and allergic group were obviously stronger than that in control group. They were mainly expressed in cytoplasm and/or nucleus of glandular cells, epithelial cells and endothelial cells, inflammatory cells. 2. The expression of TGF-β1 in non-allergic group (151.43±7.67) and in allergic group (147.51 ±6.97) were significantly stronger than that in control group (181.79±2.44, P〈0.05) . The expression of TGF-β1 in allergic group (147.51 ±6.97) was also stronger than that in non-allergic group (151.43±7.67, P〈0.05) . 3. The expression of bFGF in non-allergic group (147.33±9.86) and in allergic group (145.25 ± 7.66) were significantly stronger than that in control group (184.74±2.27, P〈0.05) . There were no statistical significance between the expression of bFGF in allergic group (145.25±7.66) and non-allergic group (147.33±9.86, P〉0.05) . 4. There were positive relationship between the expression of TGF-β1 and bFGF in both non-allergic group (r=0.364, P〈0.05) and allergic group (r=0.442, P〈0.05) . CONCLUSION 1.
