角蛋白16在喉癌中的表达及临床意义

摘　　要：目的探讨角蛋白16(KRT16)在喉癌中的作用及临床意义。方法应用实时荧光定量PCR对KRT16基因在20例喉癌及癌旁正常黏膜组织中基因水平进行定量检测,同时制作组织芯片应用免疫组化方法检测其在50例喉癌及癌旁正常黏膜组织中蛋白水平的表达。结果喉癌组及癌旁正常黏膜组KRT16 mRNA的相对表达量分别为0.732±0.201和0.201±0.134,KRT16蛋白的阳性率分别为84.0%(42/50)和22.2%(10/45),两组比较差异均有统计学意义;KRT16的表达与与患者性别、年龄无显著相关性,而与TNM分期、淋巴结转移有显著相关。结论KRT16在喉癌中的表达与肿瘤的发生、侵袭和转移密切相关,可能成为喉癌新的肿瘤标志物或预后因子。OBJECTIVE To investigate the expression of KRT16 in laryngeal squamous cell carcinoma （LSCC） and their relationship with clinicopathological features. METHODS The mRNA expression of KRT16 was determined by real-time quantitative PCR in 20 laryngeal carcinoma tissues and para-carcinoma normal tissues.lmmunohistochemistry was used to quantitatively analyze the protein levels with high-quality tissue microarray （TMA） in 50 laryngeal carcinoma tissues and para-carcinoma normal tissues. RESULTS The mRNA expression of KRT16 in LSCC tissues and the para-carcinoma normal tissues were 0.732±0.201 and 0.201±0.134 respectively （P〈0.05） . The positive expression rate of KRT16 protein in LSCC tissues and the para-carcinoma normal tissues were 84.0% （42/50） and 22.2% respectively （10/45, P〈0.05） . There were no significant correlation between the expression of KRT16 and the gender, age of the patients, but related to TNM stage and lymph node metastases. CONCLUSION Based on these findings, overexpression of KRT16 in LSCC suggested that it may participate the genesis and development of LSCC. KRT16 could be a useful tumor marker and prognostic factor for LSCC.
