摘  要:目的观察不同频率电针对帕金森病(PD)模型大鼠腹侧被盖区(VTA)酪氨酸羟化酶(TH)和神经元型一氧化氮合酶(nNOS)表达的影响。方法将30只SD大鼠,随机分为5组:正常对照组,假手术组、模型组,PD模型低频电针组和高频电针组。采用右侧纹状体内注射6-羟基多巴胺(6-OHDA)制备PD模型,取合谷和太冲穴,分别给予低频(2Hz)和高频(100Hz)电针治疗。免疫组织化学方法观察VTA的TH和nNOS表达。结果与正常对照组相比,PD模型大鼠VTA的TH表达减少、nNOS表达增加,高频电针可增加其TH表达和降低nNOS表达,低频电针对其没有影响。结论高频电针治疗PD的机制之一可能是通过降低PD模型大鼠VTA nNOS表达,从而减少因NO的产生引起的TH标记的DA能神经元的死亡。Objective To observe the expressions of tyrosine hydroxylase (TH) and neuronal nitric oxide synthase (nNOS) in the ventral tegmental area (VTA) of the Parkinson's disease (PD) model rats by electroacupuncture (EA) of different frequencies. Methods Thirty Sprague-Dawley rats were randomly divided into 5 groups:normal control, sham-operated, model, low-frequency EA treatment, and high- frequency EA treatment. PD model was prepared by injection of 6- hydroxyl-dopamine (6-OHDA) into the right striatum. PD model rats of EA groups, were given low-frequency (2 Hz) and high-frequency (100 Hz) EA treatment to the acupoints of "Hegu" and "Taichong'. Immuno-histochemistry was used to ob- serve the TH and nNOS expressions. Results Compared with those of the control group, TH expression was reduced and nNOS expression increased in VTA of PD model rats. High-frequency EA improved its TH expression and decreased nNOS expression, while low-frequency EA had no effect. Conclusion One of the mechanisms of high-frequency EA treatment of PD may be to decrease nNOS expression in VTA , thereby reducing neuronal death of TH-laheled DA caused by the production of NO.
