摘  要:目的:探讨五子衍宗丸含药血清对大鼠睾丸支持细胞分泌产物的影响及其作用机制。方法:制备五子衍宗丸含药血清,分离培养大鼠睾丸支持细胞,构建Cox7a2重组质粒,将Cox7a2重组质粒转染的支持细胞和五子衍宗丸含药血清共孵育,收集细胞上清液,采用ELISA法测定雄激素结合蛋白(ABP)、转铁蛋白(TF)、抑制素B(INHB)、IL-1β和IL-6含量。结果:与正常细胞组相比,转染Cox7a2重组质粒后支持细胞上清液中ABP、IL-1β、IL-6水平显著升高,TF、INHB显著降低(P<0.05);给予五子衍宗丸高浓度含药血清共孵育后,转染重组质粒后升高的ABP、IL-1β和IL-6水平显著降低,而降低的TF、INHB水平显著升高(P<0.05)。结论:Cox7a2的过表达可干扰支持细胞分泌功能,而五子衍宗丸含药血清对支持细胞分泌产物的改善作用与抑制Cox7a2的表达有关。Objective: To investigate the effect of Wuziyanzong Pill medicated serum on secretion of rat sertoli cell (SC)and mechanism. Methods: Primary purified rat SC was cultured and transfected with Cox7a2 recombint plasmid. After transfection, SC were cultured with Wuziyanzong Pill medicated serum for 24 hours. Cell culture supernatants were collected and the levels of androgen binding protein (ABP), transferrin (TF), inhibin B (INHB), interleukin (IL)-113 and IL-6 in culture medium were quantified by ELISA. Results: Compared with normal SC group, the level of ABP, IL-113 and IL-6 increased and the level of TF and INHB decreased significantlyin Cox7a2 recombint plasmid group. When high concentration Wuziyanzong Pill medicated serum was used, Cox7a2 recombint plasmid induction of ABP, IL-113 and IL-6 and reduction of TF and INHB were prevented. Conclusion: Cox7a2 overexpression disrupted rat SC secretion and improved effect on SC secretion by Wuziyanzong Pill medicated serum are partially mediated by inhibiting Cox7a2 expression in SC.
