摘 要:目的:探讨天麻对缺氧诱导大鼠大脑皮质神经元细胞凋亡的保护作用。方法:制备体外培养大鼠大脑皮层神经细胞缺氧模型,应用包括倒置显微镜下观察正常和缺氧神经细胞的生长情况、乳酸脱氢酶(LDH)释放率测定、流式细胞技术、TUNEL法的方法观察天麻对缺氧神经元细胞凋亡的保护作用。结果:缺氧处理后细胞LDH释放率测定显示,天麻各剂量组与尼莫地平组均可降低LDH的释放率,天麻低剂量组与尼莫地平组之间相比有明显差异(P<0.05)。流式细胞仪凋亡细胞记数:在缺氧处理前用天麻和尼莫地平预处理均可减少细胞的凋亡,天麻低剂量组与尼莫地平组之间相比有明显差异(P<0.05)。TUNEL法观察:在缺氧处理前用天麻和尼莫地平预处理均可抑制TUNEL阳性细胞的表达,天麻低剂量组与尼莫地平组之间相比有明显差异(P<0.05)。结论:天麻可明显减轻缺氧引起的大鼠大脑皮质神经细胞凋亡的发生,减轻细胞损伤,从而达到对缺氧神经细胞的保护作用。Objective: To investigate the protective effect of Gastrodia tuer on the rat corticocerebral apoptosis induced by hypoxia. Methods: Preparation the anoxic model of rat cerebral cortex nerve cells ouside of the body. And the rats were divided into normal control group, bypoxia model group, gastrodin intervention group, nimodipine control group. Then a series of indicators were selected, including the growth situation of the normal and anoxic neuron, the determination of the lactate dehydrogenase release, flow cytometry, and observation by TUNEL on the neuronal apoptosis in hypoxic changes after gastrodin intervention. Results: The determination of the rate of lactate dehydrogenase release showed that the gastrodina intervention groups and the nimodipine group all could reduce the ~;ate of the lactate dehydrogenase release rate. Compared with the nimodipine group, the Gastrodiae low-dose group showed significantly different (P〈0.05), but Gastrodiae higher-dose group and the nimodipine group didn't show significantly different. Counting the apoptotic cells by flow cytometry: pretreatment the anoxia by intervention of gastrodin and nimodipine could reduce apoptosis, the Gastrodiae low-dose group and the nimodipine group showed significantly different (P〈0.05), but Gastrodiae higher-dose group and the nimodipine group didn't show significantly different. The TUNEL observation: pretreatment the anoxia by intervention of gastrodin and nimodipine could inhibit the expression of TUNEL-positive cells, the Gastrodiae low-dose group and the nimodipine group showed significantly different (P〈0.05), but Gastrodiae higherdose group and the nimodipine group didn't show significantly different. Conclusion: Gastrodia could obviously reduce cell injury and rat cerebral cortical nerve cells apoptosis induced by hypoxia, so as to protect the nerve cells apoptosis induced by hypoxia.
