摘 要:目的:观察音乐电针和脉冲电针对慢性应激抑郁模型大鼠行为学和海马星形胶质细胞(As)的影响,探讨电针抗抑郁作用的生物学机制,并比较两种电针的疗效。方法:采用慢性应激刺激结合孤养的方式建立大鼠慢性应激抑郁模型,分组后分别采用电针、音乐电针、氟西汀治疗21d,通过旷场实验观察大鼠的行为学变化;应用As标记物——胶质纤维酸性蛋白(GFAP)免疫组织化学染色观察As形态改变及GFAP蛋白表达情况;用实时荧光定量PCR法检测各组大鼠海马内GFAP mRNA表达量。结果:模型组大鼠旷场实验水平运动得分和垂直运动得分明显降低,音乐电针、脉冲电针和氟西汀可逆转此变化;模型组大鼠海马As的数量和突起数量明显降低,GFAP蛋白和mRNA表达均明显下降,音乐电针、脉冲电针和氟西汀可改善此病理变化,并且音乐电针、脉冲电针在上调GFAP表达方面明显优于氟西汀。结论:音乐电针和脉冲电针可通过增加海马内GFAP表达、促进As增殖而发挥抗抑郁作用;在改善慢性应激抑郁大鼠的行为学及GFAP表达方面,音乐电针有优于脉冲电针的趋势。Objective: To observe the influence of music electro-acupuncture and pulsed electro-acupuncture to the locomotor activity and hippocampus astrocyte of chronic unpredictable mild stress depression model rats. Probe into the biological mechanisms of electro-acupuncture treating depression and compare the therapeutic effects of these two kinds of electro- acupuncture. Methods: Use the chronic unpredictable mild stress combined with solitarily feeding method to make the depression model. Observe the locomotor activity of rats by open-field experiment. Observe the shape of astrocyte and Glial fabrilary acidic protein by using immunohistochemistry staining GFAP. Detect the GFAP mRNA in the hippocampus of each rats by real- time PCR. Results: The cross and rear score of the model group are significantly lower than blank group while music electro-acupuncture, pulsed electro-acupuncture and fluoxetine could reverse it. The immunohistochemistry shows that the amount of the astrocyte and its bulge, the GFAP protein and mRNA are all significantly lower, the three kinds of treatment can improve these pathological change, and music electro-acupuncture and pulsed electro-acupuncture are better than fluoxetine in up-regulating the expression of GFAP protein. Conclusion: Music electro-acupuncture and pulsed electro-acupuncture could treat depression by increasing the GFAP and astrocyte in the hippocampus. In the aspect of improving the locomotor activity and GFAP of depression rats, music electro-acupuncture is better than pulsed electro-acupuncture.
