摘 要:目的:研究贝母地上部位提取物的提取纯化工艺,确定工艺参数。方法:以总生物碱为考察指标,采用酸性染料比色法测定总生物碱含量,对影响总生物碱提取率的各因素分别进行单因素考察,并用响应面法对其进行优化试验。结果:用贝母地上部位25倍量的75%乙醇(v/v)浸泡1.5h后,85℃水浴回流提取2次,每次1.0h为贝母地上部分总生物碱最佳提取条件。结论:本研究提取方法可作为贝母地上部位提取物提取工艺的一种最佳提取方法。Objective: To optimize the extraction and purification technology of the above-ground part of Fritillaria Wabuensis's extract, and confirm the technological parameters. Methods: By acidic dyes colorimetric determination of total alkaloid content, we have the single factor of investigation respectively to every factor that affect the extraction yield of total alkaloid and to optimize text by the response surface method with total alkaloid content. Results: When the extracting process was 2 hour, 2 times and 25 times weight of the material powder with 85%EtOH(v/v), the sample extract had the highest rate of extraction. Conclution: The technology prosess of this experiment can be used as a good selection of the extraction methods of above-ground part of Fritillaria Wabuensis's extract.
