摘 要:目的:研究中华眼镜蛇毒组分C(FC)对体外培养的小鼠B16黑色素瘤细胞生长的抑制作用。方法:采用MTT比色法、生长抑制实验、集落形成实验,探讨FC对小鼠B16黑色素瘤细胞株的毒性作用。结果:FC对B16细胞有明显的毒性作用,处理细胞48h,其IC50=1.75μg/ml。FC处理B16细胞24、48、72h后,各时间点的细胞存活数随FC的浓度升高而减少,细胞的生长受到抑制。FC还显著抑制B16细胞的集落形成,当FC浓度为0.5、1.0、2.0μg/ml时,其抑制率分别为33.42%、50.57%、77.34%。结论:中华眼镜蛇毒组分C对B16细胞具有明显的细胞毒性和生长抑制作用。Objective:To verify the inhibitory effects of fraction C (FC) extracted from Naja naja actra venom on growth of B16 melanoma cells. Methods:Studies were performed by means of MTT colorimetric,trypan blue exclusion and colony formation methods.Results:FC had obvious cytotoxicity on B16 melanoma cells,and IC50 was 1.75 μg/ml when B16 cells were exposed to FC for 48 h.When B16 cells were exposed to FC for 24,48,72 hours respectively,the cell survival rate decreased with the increase of FC concentration.FC also possessed inhibitory effect on colony formation of B16 cells, IR reached 33.42%,50.57% and 77.34% respectively when concentration of FC was 0.5,1.0 and 2.0 μg/ml.Conclusion:FC possesses remarkable cytotoxicity and inhibitory effect on growth of B16 cells.
