摘　　要：研制了细胞生理多参数自动分析仪的软件系统.该软件系统结合了多参数检测的细胞传感器集成芯片和自动分析控制仪器,可以实时检测和辨识细胞的多种生理参数.系统实时动态长时程采集、记录和分析了细胞的电生理活动、细胞的代谢参数以及细胞的生长状态,该软件系统具有显示界面灵活、操作界面方便以及数据的自动传输等功能;此外,研究了基于细胞生理多参数检测的自动识别算法,包括细胞电位的峰值自动检测辨识、细胞数量和抑制细胞生长药物浓度的自动判别、细胞外代谢离子成分的定量检测.使用该软件系统对实际药物进行了检测实验,给出了阿霉素作用于大鼠心肌细胞的半数抑制浓度曲线.分析了自动工作点检测算法的参数选取范围,确定了最佳测量间隔,此时的误差为13.5±1.9%.自动峰值检测算法在3种不同信噪比条件下的准确率均高于90%.A software system for automatic multifunctional cell physiological analyzer was designed for realizing automatic detection and analysis of cell physiological parameters. The system which combined with an integrated chip of cell-based biosensors and the cell physiological analyzer was utilized to detect a variety of physiological parameters such as extracellular ion concentrations, extracellular action potentials and electrophysiological impedance. A series of intelligent algorithms were adopted to analyze the cell electrophysiology, including the cell potential with peak detection, changes of cell impedance with an algorithm to dynamically calculate the half maximal inhibitory concentration, and extracellular ions parameters with operating points identification on I-V curves. After using the software system for drug testing, the half maximal inhibitory concentration curve of doxorubicin in rat cardiac myocytes was showed by the result, and of the algorithm parameters for automatic operating point detection were determined with a 13.54-1.9% error range. The automatic peak detection algorithm was utilized in three groups of different signal-to-noise rates with over 90% accuracy.
