摘 要:目的探讨红芪多糖-3对实验性2型糖尿病(T_2DM)大鼠脂质过氧化作用和对胸腺指数、脾脏指数的影响。方法建立实验性T_2DM大鼠模型,按体重随机分为模型组、红芪多糖-3大剂量组(200 mg/(kg·d))、中剂量组(100mg/(kg·d))、小剂量组(50 mg/(kg·d))及二甲双胍组(200 mg/(kg·d)),各组均灌胃给药。给药5周后取血清测定血糖、游离脂肪酸、总抗氧化能力、超氧化物歧化酶、丙二醛、一氧化氮和一氧化氮合酶的含量;取免疫器官脾脏和胸腺称重,计算内脏指数。结果与模型组比较,红芪多糖-3各剂量组均可降低T_2DM大鼠空腹血糖;提高一氧化氮合酶和超氧化物歧化酶的活性及总抗氧化能力,促进一氧化氮产生;降低血清中丙二醛和游离脂肪酸的含量;红芪多糖-3能提高大鼠的脾脏指数和胸腺指数。结论红芪多糖-3能有效降低血糖,减轻糖尿病对实验大鼠机体免疫器官的损伤,增强机体抗氧化防御能力。Objective To study the effect of hedysari polysacchcride-3(HPS-3)on the lipid peroxidation and index of thymus,spleen on experimental type-2 diabetic rats.Methods The rats were selected and randomly separated into normal control,model group,type-2 diabetic model group, HPS-3(50,100,200 mg/(kg·d))different dose group and metformin(200 mg/(kg·d))treated group according to their weight to establish an experimental type-2 diabetic rats model.Rats of all groups were treated by intragastric administration.After respectively administered for 5 weeks, the contents of glucose,free fat acids,superoxide dismutase,malondialdehyde,nitric oxide and nitric oxide synthase and total antioxidant capacity in the blood serum were measured.Immune organs,spleen and thymus obtained were weighed to calculate internal organs index.Results Compared with the model group,HPS-3 of the three treated groups could reduce fasting blood glucose in experimental type-2 diabetic rats,increase nitric oxide synthase and superoxide dismutase activity and total antioxidant capacity,promote the production of nitric oxide,and decrease malondialdehyde and free fatty acids in the serum.At the same time,it may increase the rats spleen index and thymus index.Conclusion HPS-3 can lower the level of blood glucose effectively, lighten immune organ damage in diabetic rats and enhance the body antioxidant defense capability.
