摘 要:棉花GhFTL1是开花相关的调控基因。为了解该基因的表达模式,通过染色体步移的方法从棉花基因组中分离到长度约为715bp的启动子片段。结果显示:该片段在起始密码子104bp处有1个典型的TATA box,预测的转录起始位点在起始密码子前69bp处,含有与光调节有关的元件,GT-1和SP1,另外存在逆境胁迫相关等的顺式作用元件。拟南芥的FT基因启动子上有CArG box,该基因可以促进植物早花,长春花TDC基因启动子上有与光调节有关的GT-1元件,分析此序列和拟南芥的FT、长春花的TDC等基因上的一些调节元件,发现它们有很大的相似性。结论:初步推断此序列可能是GhFTL1基因的启动子片段。The cotton GhFTL1 gene is related to flowering regulation. In order to understand the gene expression pattern, the GhFTL1 promoter was obtained from using Genome Walking method from cotton genomie DNA. There existed in the sequence a typical TATA-box at 104 (from the initiation codon) ,some regulatory elements involved in light response, such as GT-1 and Spl ,and stress-related cis-acting elements. FT gene promoter in Arabidopsis thaliana has CArC_r-box, which can promote plant early flowering. TDC gene promoter in Catharantus roseus has GT-1 ,which is involved in light response. The Analysis of regu- latory elements from this sequence,Arabidopsis thaliana FT gene and Catharantus roseus TDC gene,indicates that they have a great deal of similarity. Thus,it could be preliminarily concluded that,the sequence is the promoter of GhFTL1.
