摘  要:为了明确侵染新疆椒样薄荷的病毒种类,从伊犁采集了2株表现明显病毒症状的病株。通过dsRNA分离、单引物扩增、克隆和测序,从椒样薄荷病样中获得352bp核苷酸序列。序列分析结果表明,该片段为黄瓜花叶病毒(CMV)1a基因的部分序列。根据所获病毒信息,针对CMV 2a基因、2b基因和CP基因设计了3对特异引物,对病株进行了相应目标基因的RT-PCR,结果均扩增到预期的目标片段。对其中1个目标片段进行克隆和测序,获得了包含CMV CP基因在内的774bp核苷酸序列。该研究结果表明,新疆椒样薄荷受到CMV侵染。Field samples showing red and mosaic symptoms were collected from Yili in order to determine virus infecting Mentha piperita L.. Double strand RNA (dsRNA) was extracted from leaves. A modified Single Primer Amplification Technique (SPAT) was adopted to obtain the cDNA of dsRNA, followed by cloning and sequencing. The sequence was determined to be 389 nucleotides. Sequence analysis revealed that it was partial la gene of Cucumber mosaic virus (CMV). Three anticipated gene fragments were amplified with three primer pairs designed on the basis of sequences of CMV 2a gene,2b gene and CP gene, re- spectively. One of the gene fragments was cloned and sequenced,and we obtained 774 nucleotides sequence containing CMV CP gene. It was concluded that CMV infected M. piperita L..
