摘 要:基于电渗析技术,制作了一种"三明治"式的膜分离微流控芯片,用于脱氧核糖核酸(DNA)的无损伤在线富集。以花菁类染料为荧光指示剂,实现DNA在此芯片中的在线富集观测;分别以λ-DNA和λ-DNA-血红蛋白混合体系为模型化合物,探究其在此芯片中的富集特性;对浓缩液进行凝胶电泳实验,以考察DNA是否变性,并进行聚合酶链反应,以证明本芯片回收的DNA可供进一步的生物分析。结果表明:以花菁类染料为荧光指示剂,本芯片成功实现DNA的在线富集观测;本芯片能够无损伤捕获并浓缩单一体系和简单混合体系中的DNA分子,DNA的富集倍数近50倍,回收率约30%,浓缩液可用于进一步的生物分析。Based on the technology of electrodialysis, a new microchip with membrane separation like "sandwich" was make for the non-damage enrichment of DNA online. First, by using genefinder, a cyanine dye, as fluorescence indicator, the enrichment of DNA was monitored in the microchip. Then A-DNA and A- DNA-hemoglobin, as model compounds, were separately used to study the characteristic of capturing and enriching DNA in this chip. Final, gel electrophoresis was done to study if DNA was denaturation and polymerase chain reaction (PCR) was done to prove the concentrate of DNA enriched can be used to the further bio-analysis. The results were as follows: by using genefinder, a cyanine dye, as fluorescence indicator, on-line enrichment of DNA in the microchip were monitored successfully, the chip could be used to capture and enrich DNA scathelessly from single and simple mix system, the concentrate mutiple was accessible to 50, and the recovery efficiency was about 30% , concentration can be used for further biological analysis.
