摘 要:【目的】比较接触性超敏反应(CHS)小鼠不同部位CD4+/CD8+T细胞以及辅助性T细胞(Th1/Th2)、细胞毒性T细胞(Tc1/Tc2)的平衡状态。【方法】以二硝基氟苯(DNFB)致敏和攻击BALB/c小鼠,建立CHS模型。采用三色流式细胞术分析CHS小鼠脾脏(SP)、耳后淋巴结(RLN)、小肠派氏结(PPs)和肠系膜淋巴结(MLN)中CD4+T细胞和CD8+T细胞的变化及其胞内细胞因子干扰素-γ(IFN-γ)与白细胞介素-4(IL-4)的表达水平,观察CHS小鼠不同部位Th1/Th2、Tc1/Tc2比例的变化。【结果】与正常组比较,CHS组RLN与SP中CD4/CD8比值显著下降(均P<0.01),RLN中以Tc1与Th1占优势,SP中以Tc1与Th2占优势,PPs中Tc1细胞比例、CD4/CD8比值显著上升(均P<0.05),MLN中各指标无显著性差异。【结论】DNFB诱发的CHS小鼠机体总体免疫平衡向Th1类偏移,Tc1(CD8+IFN-γ+)细胞是主要的效应细胞。Objective To compare the changes of CD4 +/CD8 + T cell subsets and intracellular cytokines of Thl/ Th2, Tcl/Tc2 cells in different tissues of contact hypersensitivity (CHS) mice. Methods BALB/c mice were sensitized and challenged with dinitrofluorobenzene (DNFB) to induce CHS model. Trichromatism flow cytometry was adopted to detect the changes of CD4+ and CD8 + T cell subsets in the spleen ( SP), retroauricular lymph nodes ( RLN), intestinal Peyer' s patches (PPs) and mesenteric lymph nodes (MLN) as well as the expression of intracellular cytokines of interferon gamma (IFN--γ) and interleukin 4 (IL-4) cells. The changes of Thl/Th2 and Tcl/Tc2 ratio in different tissues of CHS mice were also observed. Results Compared to the normal group, CD4/ CD8 ratio decreased significantly in RLN and SP of CHS mice (P 〈0. 01 ). Tcl and Thl cells were dominated in RLN of CHS mice, Tcl and Th2 cells dominated in SP, Tcl cells percentage and CD4/CD8 ratio significantly increased in PPs ( P 〈 0. 05 ). There was no significant difference in the indexes of MLN. Conclusion The murine contact hypersensitivity is mainly mediated by Tcl ( CD8 + IFN-γ+ ) effector cells, and the intestinal mucosal immunity and system immunity in DNFB-induced CHS mice are dominated by Thl cells.
