Effect of Traumatic Cataplasma on Aquaporin Protein-3 Expression in Soft Tissue of Traumatic Injury Rat Model

摘  要:【目的】观察治伤巴布剂(血壳、虎杖等)对大鼠创伤性软组织肿胀模型骨骼肌组织中水通道蛋白-3(AQP-3)表达的影响。【方法】选用SD大鼠54只,随机分成3组,造模前1 d使用100 g/L硫化钠对每只大鼠左后肢大腿外侧区域进行脱毛处理。正常对照组仅在左后肢大腿外侧进行脱毛处理,并标记打击范围,不予打击造模;模型组使用软组织打击器在脱毛区标记范围内予以打击复制急性软组织损伤肿胀的模型,不给任何药物处理;药物处理组于急性软组织损伤肿胀模型建立后立即在局部给予治伤巴布剂外敷,另给予胶布加强固定。分别于造模后1 h、6 h、1 d、3 d、5 d、7 d 6个时相点处死动物,每组每个时相点3只,在标记的区域内肌肉组织取材,采用干湿比重法测定骨骼肌组织的含水量,应用WesternBlot技术检测AQP-3蛋白表达水平。【结果】模型组的肌肉组织含水量均显著高于正常对照组(P<0.05),在3 d、5 d、7 d3个时相点,药物处理组的含水量显著低于模型组(P<0.01);模型组AQP-3蛋白表达水平均显著高于正常对照组(P<0.01),而药物处理组AQP表达水平显著升高,与模型组比较差异具有统计学意义(P<0.01)。【结论】治伤巴布剂减轻急性软组织肿胀从而加速急性软组织损伤后修复的作用可能与其能上调APQ-3表达水平有关。Objective To observe the effect of Traumatic Cataplasma on the expression of aquaporin protein-3 ( AQP-3 ) in skeletal muscle of rat model with soft tissue swelling induced by traumatic injury. Methods Fifty-four SD rats were randomized into normal control group, model group and treatment group. One day before the modeling, depilating treatment with 100g/L sodium sulfide was made in the lateral region of rat left thigh. Normal control group was given depilating treatment, and then the striking range was marked in the depilatory area, but the striking was not carried out. Model group was given the striking in the depilatory area with the blowing apparatus for establishing the model of soft tissue swelling induced by acute injury. The treatment group was immediately applied Traumatic Cataplasma externally on the swelling area, and fixed with bandage after establishment of the model. Three rats were executed in each group at time point of modeling hour 1 and 6, and at modeling day 1, 3 , 5 and 7 d, respectively. Muscle tissue was sampled in the marked area of rats, water content of the skeletal muscle was detected after calculating the proportion of dry weight and wet weight, and the Western Blot assay was applied for the examination of AQP-3 protein expression level. Results At all of the six time points, the model group had higher water content in skeletal muscle than the normal control group ( P 〈 0.05 ). At time point of modeling day 3, 5 and 7, treatment group had lower water content than the model group (P 〈 0. 01 ). Model group had higher AQP-3 protein expression level than the normal control group ( P 〈 0. 01 ), and the treatment group had higher expression level than the model group ( P 〈 0.01 ). Conclusion Traumatic Cataplasma could relieve soft tissue swelling induced by acute injury , and accelerate the repair of the injured soft tissue. Its possible mechanism is related with the down-regulation of AQP-3 protein expression level .
