Ghrelin重组质粒的表达及其对大鼠生长性能和胃重的影响

摘　　要：旨在研究Ghrelin重组表达质粒对大鼠生长性能和胃重的影响。从断奶仔猪胃底黏膜中扩增得到了Ghrelin,将其克隆到pcDNA3表达载体中,获得了重组质粒pcDNA3-Ghrelin。选取12只断乳大鼠随机分成2组,每组6只。试验组每只大鼠腿肌注射100μg重组质粒pcDNA3-Ghrelin,对照组注射空质粒。分别在注射后7 d、14 d和29 d称重、记录采食量。试验结束后处死,分离并称量胃重,测定胃液pH值。结果显示:与对照组相比,注射重组质粒大鼠在注射后7 d和29 d时日增重显著增高;采食量无显著差异;料肉比显著降低;胃重和相对质量无显著差异;胃液pH值显著降低。以上结果表明,Ghrelin通过肌肉组织转染表达后对大鼠的生长发育具有重要的调节作用。The experiment aimed to study the effects of recombinant Chrelin plasmld on growth performance and stomach weight of rats. Ghrelin coding region was amplified from the gastric fundus mucosa of weaned piglets and cloned into the vector pcDNA3 to construct an expression vector named pcDNA3-Ghrelin. Twelve weaned rats were divided randomly into two groups. 100 μg pcDNA3-Ghrelin was injected intramuscularly into rats as treatment, and 100 μg empty plasmid as control. Body weight and feed consumption were recorded on 7 d, 14 d and 29 d after injection, respectively. The rats were killed at 29 d to get weight and pH value of stomach. Compared to control, daily gain of rat in treatment group was increased on 7 d and 29 d after injection, feed consumption was not significantly different, feed conversion ratio was reduced, stomach weight and relative index （stomach weigh/body weight） was not significantly different, and stomach pH value declined. These results suggested that intramuscular injection of pcDNA3-Ghrelin could improve growth development of weaned rats.
