肾血管性高血压大鼠胸主动脉内中介素IMD基因与RAAS的相关性研究

摘  要:目的:研究肾血管性高血压大鼠肥厚的胸主动脉内中介素(IMD)及受体的变化与RAAS的激活是否有关系。方法:将体重280~320 g雄性SD大鼠不完全结扎一侧肾动脉,结扎后4周测量血压,以收缩压>140 mm Hg(1 mm Hg=0.1333 kPa)为标准认为造模成功,并随机分成4组:肾血管性高血压组(Rh,n=8),结扎加缬沙坦(Val,n=7)组,结扎加左旋氨氯地平组(Aml,n=7)和结扎加依那普利组(Ena,n=9),对照组进行假手术(So,n=6)。每2周测量一次体重、血压并调整用药剂量至满10周。尾套法测收缩压(SBP),放射免疫法检测胸主动脉内血管紧张素Ⅱ(AngⅡ)和醛固酮(ALD)含量,RT-PCR法测定胸主动脉内IMDmRNA、RAMP1、2、3mRNA和CRLR mRNA。运用SPOT VERSION 4.6显微图像分析软件测定胸主动脉中膜层厚度(MT)及中膜截面积(MA)。结果:SBP和MT、MA在Rh组较其他组明显升高,其余组间差异无统计学意义(P>0.05)。实验前、后各组心率无变化。ALD含量,Rh、Aml组显著高于其他三组,Ena、Val组又显著高于So组。AngⅡ含量Rh组、Val与Aml两两比较,差异无统计学意义(P>0.05),且显著高于Ena组与So组。IMDmRNA在Rh组相应地显著高于其他四组,在其他各组间差异无统计学意义(P>0.05)。结论:IMDmRNA基因表达可能参与肾血管性高血压大鼠胸主动脉肥厚的病理生理过程,但与胸主动脉局部RAAS无关。Objective To investigate the expression of intermedin ( IMD), including the ligand and its receptors in the hypertrophied thoracic aorta and the relation between the change and RAAS in renovascular hypertensive rats. Methods male SD rats weighing280 - 320 g were chose for establishment of hypertensive animal model by incomplete ligation of the left renal artery. The successfully made rats (SBP 〉 140 mm Hg)were randomly divided into renovascular hypertension Rh group( Rh group, n = 8 )and treatment groups (Val group n = 7, Am1 group n = 7, Ena group n = 9), which were administered with Valsartan, Amlodipine and Enalapril respectively. A sham - opera- tion group (So group, n = 6)was set up as negative control. The systolic blood pressure (SBP)was measured by tail cuff, and media thickness (MT) and media area(MA) of thoracic aorta were determined in the t0th week after drug interventiont, and the level of Angiotensin II( Ang II) and aldosterone(ALD) in myocardium were measured by radioimmunoassav. Meanwhile,gene expression of IMDmRNA was detected by RT - PCR. Results SBP,MT and MA were increased in Rh compared with those in the other groups respectively. There are no significant difference in heart rate at 4 weeks after operation in all groups. The levels of ALD concentration in thoracica aortic in Rh and Aml group were significantly higher than those in the other groups, while Ena and Val group are significantly higher than So groups. There was no a sig- nificant difference in the level of Ang 1~ concentration in thoracica aortic among Rh, Val and Anal group by pariwise comparison, and theirs the level of Ang II were significantly higher those in the Ena and So group. The level of IMDmRNA in the thoracic aorta in Rh group were up - regulated compared with that in the other gToups correspondently. Conclusion The gene expression of IMDmRNA may take part in the pathological and physiological process of hypertrophy of thoracic aorta,but it hardly seems to be connected with the
