摘 要:非特异性脂质转移蛋白(nsLTP)是植物中大量存在的小分子脂类结合蛋白,具有抗菌防御功能。分析黄瓜非特异性脂质转运蛋白基因nsLTP的序列特征及其在细菌性角斑病菌浸染过程中的表达模式,对其应用于黄瓜等农作物转基因工程,增强作物抗病性具有重要意义。在黄瓜叶cDNA文库中,获得非特异性脂质转移蛋白(nsLTP)cDNA序列,命名为CsnsLTP;该基因全长566 bp,编码121个氨基酸残基;序列分析表明,该基因具有N端信号肽,具有nsLTP家族共有的典型保守区域,属nsLTP家族基因。实时荧光定量PCR分析表明,CsnsLTP在黄瓜叶中有表达。在黄瓜细菌性角斑病菌浸染下,该基因在黄瓜叶中表达增高,并随浸染时间的延长而增强,明显受黄瓜细菌性角斑病菌的诱导,推测该基因在抵御黄瓜细菌性角斑病菌浸染时具有重要作用。Plant non-specific lipid transfer proteins (nsLTPs) are abundantlipid binding proteins with small molecules that play several biological roles including antimicrobial defense. Analyzing the sequence characteristics of non-specific lipid transfer protein genes and their expression patterns challenged by Pseudomonas syringae pv. Lachrymans, which provides necessary data for further investiclation of the crops transqene project and disease, resistance of cucumber. The cDNA sequenceof the cucumber non-specific lipid transfer protein was obtained from the cucumber leaf cDNA library, termed as CsnsLTP; The length of CsnsLTP cDNA is 566 bp encoding 121 amino acid. sequence analysis revealed that the gene has the N terminal signal peptide and a typical nsLTP family conserved region, revealed the CsnsLTP belongs to nsLTP family gene. The expression analyses of the gene by real-time fluorescence quantitative PCR showed that the CsnsLTP express in the cucumber leaves. Challenged by Pseudomonas syringae pv. Lachrymans, the expression of the gene in cucumber leaves significantly increased with the extension of challenging time, which indicated that the CsnsLTP is induced by the challenging and might play an important role against the invasion of bacterium.
