摘 要:目的观察护卵汤对促性腺激素释放激素(GnRHa)超排卵大鼠卵巢细胞凋亡及活胎数的影响,为护卵汤辅治辅助生殖技术提供实验依据。方法模仿人类辅助生殖中的长周期,对大鼠进行GnRHa垂体降调节预处理后再用尿促性腺激素(HMG)等药物超排卵(下称模型组),同时给予不同剂量的护卵汤辅助治疗,以及自然周期未使用任何药物的大鼠为正常对照(下称正常组)。在注射绒毛膜促性腺激素(HCG)当天摘取卵巢组织用缺口末端标记法(TUNEL法)检测卵巢细胞凋亡情况;在孕12 d观察各组大鼠的妊娠胚胎数。结果 HCG日模型组卵巢体细胞凋亡较正常组显著增多,差异有统计学意义(P<0.01);护卵汤各剂量组卵巢细胞凋亡均较模型组减少,差异有统计学意义(P<0.01或<0.05);孕12 d各组妊娠大鼠的总活胎率差异无统计学意义(P>0.05)。结论 GnRHa超排卵增加大鼠卵巢细胞凋亡,对卵泡发育有不利影响;而护卵汤能减少GnRHa超排卵大鼠卵巢体细胞凋亡。Objective To observe the effects of Huluan Decoction on ovary cell apoptosis and the total live fetus of rats with GnRHa controlled ovarian hyperstimulation (COH), and to provide related experimental evidence for the effectiveness of Huluan Decoction in assisted reproductive technology (ART). Methods After GnRHa pituitary down regulation pretreatment which imitated the human long period of COH, a number of rats were separated into model group treated with HMG/HCG superovulation drugs and treatment groups treated with HMG/HCG superovulation drugs and different doses of Huluantang. The rats with natural cycle were used as control group receiv- ing no medicine. The ovary cell apoptosis was observed through TUNEL on the day of HCG injec- tion. The quantity of live fetus of rats was observed with dissection on the 12th day of gestationperiod. Results Compared with control group, the increase of ovalT cell apoptosis in model group had statistical significance(P〈0.01). Compared with model group, the decrease of o- val2~ cell apoptosis in Huluan Decoction different dose groups had statistical significance (P〈0.01 or 0.05). The total live fetus in all groups have no statistical differencas. Conclusion The Gn- RHa controlled ovarian hyperstimulation may increase the cell apoptosis of ovary and has adverse effects on follicular development in rats. Huluan Decoction can decrease the cell apoptosis of ovary. and increase live fetus of rats with GnRHa controlled ovarian hyperstimulation.
