摘  要:目的:建立人前列腺癌细胞体外药敏实验,指导临床前列腺癌化疗。方法:收集前列腺癌组织标本,进行原代培养及纯化培养,获得前列腺癌细胞。接种96孔板,给予1×PPC多西他赛(DOC)、羟基喜树碱(HCPT)、顺铂(DDP)、依托泊苷(VP-16)、阿霉素(ADM)和丝裂霉素(MMC)处理,采用CCK-8法观察上述化疗药物对前列腺癌细胞的抑制率。结果:前列腺癌细胞对于上述化疗药物的敏感性依次是:DOC>HCPT>VP-16>DDP>ADM>MMC。在本实验条件下前列腺癌细胞原代培养成功率为78.1%(50/64),纯化培养成功率为65.6%(42/64)。原代培养平均耗时(16±4.2)天(n=50),纯化培养平均耗时(53±12.6)天(n=42)。纯化培养的前列腺癌细胞对化疗药物的敏感性高于原代培养的细胞。结论:原代培养前列腺癌细胞药敏实验操作简便,成功率高,耗时短,与纯化培养药敏结果相关性好,可以用于指导前列腺癌的临床治疗。Objectives: To establish a chemosensitivity test for human prostatic cancer cells in vitro and guide clinical prostate cancer chemotherapy. Methods: Prostate cancer tissue samples were collected and primary culture and purified culture were conducted. Then the cells were harvested and inoculated in 96-well plates. Docetaxel (DOC), hydroxycamptothecin (HCPT), cisplatin (DDP), etoposide (VP-16), adriamycin (ADM) and mitomyein (MMC) were respectively added at the concentration of l xPPC. Then the inhibitory rate of the cells was detected using CCK-8 assay. Results: The order of the inhibitory potency of the drugs on prostatic cancer cells was: DOC〉HCPT〉VP-16〉DDP〉ADM〉MMC. The success rate of primary culture was 78.1% (50/64) and that of purified culture was 65.6% (42/64). The average time- consuming were 16±4.2 d (n=50) and 53±12.6 d (n=42), respectively. Purified culture prostate cancer cells were more sensitive to chemotherapeutic drugs than primary culture cells. Conclusions: Primary culture cells for prostate cancer chemosensitivity tests are simple and less time-consuming. As it is well con'elated with the results of purified culture cells, it can be used to guide clinical chemotherapy of prostate cancer.
