摘  要:目的从免疫抑制角度观察益气活血解毒中药联合树突状细胞(DC)疫苗对小鼠宫颈癌移植瘤的影响,为宫颈癌的治疗提供依据。方法将1×106个(0.2 mL)U14细胞接种于小鼠背部皮下,7 d后建立荷瘤小鼠模型,将造模成功小鼠随机分为4组,每组10只。对照组0.2 mL/只生理盐水腹腔注射;中药组0.2 mL/只中药灌胃,均每日1次,连续14 d;DC疫苗组局部注射DC疫苗5×106/只,每周1次,共注射2次;联合组给予中药及DC疫苗。观察小鼠瘤质量,采用免疫组化方法测定小鼠瘤组织中转化生长因子-β1(TGF-β1)、白细胞介素-10(IL-10)及脾脏CD4+、CD8+T细胞的百分比。结果干预14 d后,联合组、中药组及DC疫苗组的瘤质量显著低于对照组,各用药组对小鼠移植瘤生长均有抑制作用,以联合组最为明显。干预后,联合组和中药组TGF-β1、IL-10显著低于DC疫苗组及对照组(P<0.05);联合组和中药组CD4+、CD8+T细胞所占百分比显著高于DC疫苗组和对照组(P<0.05)。结论中药增强DC疫苗的抗肿瘤作用与其去除免疫微环境中免疫抑制因子有关。中药与DC疫苗的联合有望成为一种有效的免疫疗法。Objective To investigate the effect of the combination of TCM, with the function of supplementing qi and activating blood as well as detoxication, and DC vaccine on the cervical cancer xenografts of mice. Methods The xenografts were established by injecting 1 × 10^6 U14 cancer cells subcutatenously on the back of mice seven days later. Forty xenografts were divided into 4 groups (n = 10): control group {0.2 mL normal saline intraperitoneally mouse once a day for 14 days), TCM group (0.2 mL TCM by gavage per mouse once a day for 14 days), DC vaccine group (injecting 5× 10^6 DC vaccine around the tumor per mouse once a week for 14 days) and the combination group (administered TCM and DC vaccine by the way of described above). The irnmunohistochemistric staining was performed to detect TGF-β1, IL-10 in the tumor tissue and CD4+, CD8+ T cells in the spleen. Results The combination group, TCM group and DC vaccine group had significantly lower tumor weight and higher inhibition rate than the control after intervention (P 〈0.05). TGF-β1 and IL-10 signiticanfly decreased in combition group and TCM group than in DC vaccine group and control group (P〈0.05). The percentages of CD4+ and CD8+ T cells were significantly higher in TCM group and the combition group than in DC vaccine group and control group (P〈0.05). Conclusion The inhibition of TCM to tumor growth could be involved in abrogation of immune suppression. So the combination of TCM and DC vaccine probably is an effective immunity-based therapy.
