摘 要:目的观察miR-143在遗传性ob/ob肥胖小鼠与正常对照组C57/BL6J小鼠肝脏、骨骼肌中的差异表达,初步探讨miR-143在肥胖及相关脂代谢紊乱发生中的作用。方法选择16周龄的ob/ob小鼠与C57/BL6J小鼠各12只,采用实时荧光定量聚合酶链反应技术检测两组小鼠肝脏、骨骼肌中miR-143的表达水平。结果 miR-143在ob/ob小鼠肝脏中的表达水平明显高于C57/BL6J小鼠(P<0.05),但在骨骼肌中的表达水平两组无显著性差异(P>0.05)。结论 miR-143可能参与了ob/ob小鼠肝脏脂质代谢紊乱的发生。Objective To observe the different expression of miR - 143 in liver tissue and skeletal muscle of inherited ob/ob fatty mice and normal control mice (C57/BL6J), and preliminarily explore the role of miR - 143 in obesity and disorder of related lipid metabolism. Methods Twelve ob/ob mice and 12 C57/BL6J mice at 16 - week - old were selected. Real - time fluorescent quantitative polymerase chain reaction technique was used to detect the expression level of miR - 143 in the liver and skeletal muscle of those mice. Results The expression level of miR - 143 in the liver of ob/ob mice was significantly higher than that in C57/BL6J mice ( P 〈 0.05 ) , but there was no significant difference in the expression in skeletal muscle between the two groups ( P 〉 0.05 ). Conclusion MiR - 143 may participate the occurrence of liver lipid metabolism disorder in ob/ob mice.
