摘  要:目的研究乳源免疫调节肽(PGPIPN)对小鼠酒精性脂肪肝的防治作用。方法 60只昆明种雄性小鼠,随机均分为对照组、模型组、护肝片(0.3 g/kg)组、PGPIPNⅠ组、Ⅱ组和Ⅲ组。采用梯度乙醇溶液灌胃的方法建立小鼠酒精性脂肪肝(AFL)模型,造模的同时给予药物干预,共12周。实验结束后,处死小鼠,收集血清和肝脏。测定小鼠肝指数(LI)、脾指数(SI)、血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)及高密度脂蛋白(HDL-C)和肝组织中甘油三酯(TG)、丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)含量,并观察肝脏病理组织学变化。结果 PGPIPN各剂量组均能显著降低AFL小鼠LI和SI水平、血清中ALT和AST的活性,减少血清中TG、TC及LDL-C和肝组织中TG、MDA含量,升高HDL-C、SOD含量和GSH-PX活性水平;同时,病理组织学检查结果显示,PGPIPN各剂量组小鼠肝脏中小泡性脂肪变性程度明显减轻。结论PGPIPN对酒精性脂肪肝小鼠有防治作用,其机制可能与其调节脂质代谢,抗脂质过氧化有关。Objective To study the protective and therapeutic effects of immunomodulating peptide (PGPIPN) on alcoholic fatty liver (AFL) in alcohol-fed mice. Methods 60 Kunming mice were randomly divided into six groups: control, model, HGP, PGPIPN I , PGPIPN l] and PGPIPN m. Mice were drenched with liquor to make the model of AFL. The control group was given equal normal saline. The whole experiment lasted 12 weeks. At the end of the experiment, the mice were killed to collect the samples of sera and liver tissues. The liver index (LI) and spleen index (SI) were calculated. The contents of triglyceride ( TG), total cholesterol ( TC ), low density lip- oprotein C (LDL-C) and high density lipoprotein C (HDL-C), and activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were assayed in the sera. The contents of TG and malondialdehyde ( MDA), and activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) were also determined in liver homogenate. The pathological changes of tissues were observed. Results PGPIPN could reduce the levels of LI and SI, the activities of ALT and AST, and the contents of TG, TC, LDL-C. MDA in mice sera were remarkably decreased, the content of SOD, HDL-C, GSH-PX in mice sera were increased. Liver histopathological examination showed PGPIPN could ameliorate hepatic steatosis. Conclusion PGPIPN has the preventive effect on AFL in mice. PGPIPN could operate with modulation of fat metabolism and reduction of lipid peroxidase for prevention of AFL.
