摘 要:目的研究兔VX2肺移植瘤模型对18F-脱氧葡萄糖(18F-FDG)的摄取及其与葡萄糖转运蛋白(GLUT)-1、乏氧诱导因子(HIF)-1α的表达之间的相关性,探讨18F-FDG PET/CT评估肿瘤乏氧的价值。方法建立16只兔VX2肺移植瘤模型,分别于肿瘤生长第5、6、7、8周行18F-FDG PET/CT显像,采集不同时期模型兔肺部肿瘤最大标准化摄取值(SUVmax-T)、对侧正常肺组织最大标准化摄取值(SUVmax-N)及两者的比值(T/N),每次显像结束后随机处死4只模型兔,取肿瘤组织行GLUT-1及HIF-1α免疫组织化学染色,将肿瘤组织中GLUT-1、HIF-1α的表达水平及T/N值进行相关性分析。结果 4组T/N值分别为(9.11±0.62)、(12.96±0.42)、(13.93±0.80)、(11.49±0.71),肿瘤第5、6、7、8周T/N值由低到高再降低,差异有统计学意义(F=40.74,P<0.01)。GLUT-1表达与T/N值呈正相关(r=0.91,P<0.05),HIF-1α表达与T/N值呈正相关(r=0.75,P<0.05),GLUT-1表达与HIF-1α表达呈正相关(r=0.66,P<0.05)。结论18F-FDG的摄取与GLUT-1及HIF-1α的表达密切相关,18F-FDG PET/CT可对在体肿瘤乏氧状态进行动态化、定量化监测。Objective To investigate the correlation between F-FDG uptake and the expressions of GLUT1 and HIF-lα in rabbit lung VX2 transplant tumor, and to explore the value of 1SF-FDG PET/CT to assess hypoxia status in tumor. Methods 16 rabbits were implanted with VX2 tumor tissue in lung,then lSF-FDG PET/CT imaging was performed on those rabbits in the 5th,6th,7th,8th week respectively, The tumor tissue SUVmax (maximum stand- ardized uptake value) ,the contralateral lung tissue SUVmax and the ratio of the two(T/N) were monitored by lSF- FDG PET/CT in each period and recorded. Four rabbits were randomly sacrificed after PET/CT imaging, and immu- nohistochemistry staining was used to assess expressions of GLUT-1 and HIF-1 or. Correlation analysis was carried out on lSF-FDG uptake and the expressions of GLUT1 and HIF-lα. Results T/N in the four different periods was (9. 11 ±0.62 ), ( 12.96 + 0.42 ), ( 13.93 + 0.80 ), ( 11.49 _+ 0.71 ), respectively. T/N increased gradually and then decreased in the four consecutive weeks, the differences being statistically significant (F = 40.74,P 〈 0.05 ). T/N was significantly correlated with GLUT1 expression (r = 0. 91, P 〈 0. 05 ) and HIF- 1 ot expression (r = 0.75, P 〈 0.05 ) . A significant positive correlation between GLUT-1 expression and HIF- l α expression was found as well (r = 0. 66,P 〈 O. 05). Conclusion The uptake of FDG is closely correlated to expressions of GLUT 1 and HIF- 1 α, and is F-FDG PET / CT can quantitatively and chronologically monitor tumor hypoxia status.
