摘  要:目的探讨黄芪多糖(APS)对RAW264.7巨噬细胞源性泡沫细胞内胆固醇含量的影响。方法采用体外培养小鼠巨噬细胞系RAW264.7细胞,用50mg/L氧化型低密度脂蛋白(OX—LDL)诱导,建立泡沫细胞模型。用油红0染色在光镜下鉴定泡沫细胞形态及变化。再以不同浓度APS(0mg/L、10mg/L、20mg/L、50mg/L、100mg/L)作用泡沫细胞24h;以100mg/LAPS作用泡沫细胞不同时间(0h、6h、12h、24h、48h)后,酶比色法检测泡沫细胞内胆固醇的含量。结果①巨噬细胞经OX—LDL诱导48h后转化为泡沫细胞。②与对照组相比,10mg/L、20mg/L、50mg/L、100mg/L的APS作用于巨噬细胞源性泡沫细胞24h后,细胞内胆固醇含量均明显减少(P〈0.01)。③与对照组相比,100mg/L的APS作用于巨噬细胞源性泡沫细胞6h、12h、24h、48h,细胞内胆固醇含量均明显减少(P〈0.01)。结论①巨噬细胞经OX—LDL诱导后,有大量泡沫细胞形成。②APS可减少RAW264.7巨噬细胞源性泡沫细胞内胆固醇含量。Objective To study the effects of astragalus polysaccharides (APS) on cholesterol content in RAW264.7 macrophage-derived foam ceils. Methods The mouse RAW264.7 cells were induced into foam ceils by incubation with 50 mg/L oxidized low density lipoprotein(ox-LDL). Transmission light microscope(using red oil O staining technique) was applied to appraise the morphology and change of foam cells. After APS of different concentrations(0 mg/L, 10 mg/L, 20 mg/L, 50 mg/L, 100 mg/L) were incubated with RAW264.7 macrophage-derived foam cells for 24 h and 100 mg/L APS was incubated with RAW264.7 macrophage-derived foam ceils for 0 h, 6 h, 12 h, 24 h, 48 h, the cholesterol content of foam cells was measured by enzyme assay. Results (1) The mouse RAW264.7 cells could be differentiated into macrophages after 48h induction by ox-LDL. (2)Compared with the control group, treatment of RAW264.7 macrophage-derived foam cells with 10 mg/L, 20 mg/L, 50 mg/L, 100 mg/L APS for 24 h resulted in an decrease in the cholesterol content of foam ceils (P〈0.01). (3)Compared with the control group, treatment of RAW264.7 macrophage-derived foam cells with 100 mg//L APS for 12 h, 24 h, 48 h, resulted in an decrease in the cholesterol content of foam ceils (P〈0.01). Conclusion (1)The mouse RAW264.7 cells could differentiate into macrophage after 48 h induction by ox-LDL. (2)APS could decrease the cholesterol content of foam ceils.
