摘 要:目的研究青稞中β-葡聚糖对高脂膳食诱导的C57小鼠胆固醇代谢的影响。方法 C57BL/6雄性小鼠40只,采用随机数字表法,按体重分层随机分为4组,即β-葡聚糖高剂量组,β-葡聚糖低剂量组和高脂对照组以及基础对照组。观察其摄食量及体重变化情况。第11周末连续3 d收集小鼠粪便,记录干湿重并测定粪便中胆汁酸、甘油三酯、粪固醇。第12周末处死小鼠,留取血液标本测定血液相关生化指标,留取肝脏进行组织切片观察和分子生物学研究。结果干预12周后,与高脂对照组相比,高剂量组小鼠血清高密度脂蛋白胆固醇(high densitylipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、甘油三酯、总胆固醇水平均值分别降低了38.19%、70.31%、24.14%、15.53%,差异均有统计学意义(均有P<0.05),但HDL-C/LDL-C比值却高于高脂对照组。高剂量组小鼠粪便湿重均值、胆汁酸含量均值、甘油三酯含量均值、胆固醇含量均值较高脂对照组分别增加7.34%、16.07%、40.73%、113.70%。高、低剂量组小鼠肝脂肪变性较高脂对照组小鼠轻,肝脏组织甘油三酯含量均值分别降低47.36%、35.77%。同时,高剂量组小鼠肝脏CYP7A1 mRNA、LDL-R mRNA、HMG-CoAR mRNA的表达水平分别是高脂对照组的7.85倍、1.86倍、2.26倍。结论青稞β-葡聚糖具有促进胆固醇在肝脏中转换为胆汁酸并经粪便排泄,从而降低血清胆固醇水平的作用。Objective Figure out the effects of beta glucan in highland barley on cholesterol metabolism in C57 mice induced by high fat diet. Methods 40 male C57BL/6 mouse were stratified randomly divided into 4 groups with the table of random number by weight : high dosage group, low dosage group, high fat diet group and control group. Food-intake and body weight of mice were observed. Feces were collected over 3 days after 11 weeks of treatment, wet and dried weight were recorded. Concentration of fecal bile acid, sterol and triglyceride were determined by kit. Mice were euthanized after 12 weeks of trail, blood was obtained for biochemical examination and livers were collected for histological section and molecule biological study. Results After 12 weeks treatment, compared with the high fat diet group, the mean of serum high density lipoprotein cholesterol ( HDL-C ), serum low density lipoprotein cholesterol ( LDL-C ), serum triglyceride and total cholesterol of mice in high dosage group were decreased by 38. 19%, 70.31% , 24. 14% , 15.53% separately. Ratio of HDL-C to LDL- C was significantly rised. Compared to the high fat group, the mean of high dosage group had the wet weight of feces, content of bile acid, triglyceride and cholesterol were increased by 7. 34%, 16. 07%, 40. 73%, 113. 70% respectively. Both in high and low dosage group, hepatic steatosis of mice were lighter than that in mice of high fat diet group while the content of triglyceride in livers were decreased by 47. 36%, 35. 77% in the two groups. The expression levels of CYP7AlmRNA, LDL-R mRNA, HMG-CoAR mRNA in high dosage group were respectively 7. 85-fold, 1.86-fold and 2. 26-fold of the high fat control group. Conclusions Beta glucan in highland barley can promote the cholesterol in liver conversion for bile acid and the feces excretion, thus it plays a vital role in reducing the level of serum cholesterol.
