摘　　要：目的研究不同浓度白细胞介素18(interleukin-18,IL-18)对人早孕滋养细胞MMP-9和TIMP-1表达的影响。方法分离、培养早孕期滋养细胞,以不同浓度的IL-18作用于体外培养的细胞滋养细胞。PCR(Real-TimePCR)检测MMP-9及TIMP-1 mRNA的表达。结果随着IL-18浓度的增高,滋养细胞中MMP-9 mRNA的表达水平下降(P<0.01),TIMP-1 mRNA的表达水平则相反(P<0.05)。结论 IL-18能减弱滋养细胞表达MMP-9的能力,而增强TIMP-1的表达,IL-18可能通过节制性调节MMPs/TIMPs平衡,进而影响滋养细胞的侵袭。Objective To investigate the effect of IL-18 on the expression of MMP-9 and TIMP-1 in trophoblasts of human first trimester gestation. Methods The trophoblasts of human first trimester gestation were cultured and treated with different concentrations of IL-18. The expression of MMP-9 and TIMP-1 was detected by Real-Time PCR. Results The expression of MMP-9mRNA was declined significantly with the increased concentrations of IL-18, while the expression of TIMP-lmRNA was increased. Conclusion IL-18 can down-regulate the expression of MMP-9mRNA and up-regulate the expression of TIMP-1 mRNA, which may affect the invasion of trophoblasts.
