摘 要:【目的】探讨鸭源多杀性巴氏杆菌(Pasteurella multocida,PM)强毒株对雏鸭的致病性。【方法】以口服灭菌生理盐水的雏鸭为对照,通过口服和滴鼻接种途径将PM人工感染雏鸭,观察雏鸭的发病情况;于接种后不同时间内随机剖杀试验组和对照组雏鸭,采集肝脏、肺、心脏和脾脏、胸腺、腺胃和小肠等组织,常规方法制备石蜡切片,进行H.E.染色,观察各器官组织学变化;利用建立的间接免疫荧光法对PM在雏鸭体内的分布进行研究。【结果】接种PM后,引起雏鸭出现精神不振,呼吸困难,腹泻等症状;剖检病变以心包积液、肺出血和肝脏点状坏死为特征。PM动态增殖检测结果表明:滴鼻接种后12h,肺中PM含量最高,达到4.47×103 CFU/g,而口服接种后12h,胸腺中PM含量最高,达到5.12×103 CFU/g;2种途径接种PM后18~30h均以雏鸭胸腺PM含量最高。病理组织学变化以肺脏充血、出血,肝脏脂肪变性,脾索紊乱等为特征;荧光阳性信号主要分布于肺、肝脏、胸腺和脾脏等器官。【结论】PM感染主要引起雏鸭的出血、败血性变化;滴鼻接种PM对鸭的致病力明显高于口服接种;胸腺、肺、肝脏和脾脏是PM侵害的主要靶器官。[Objective] This paper aimed to study the pathogenicity of Pasteurella multocida (PM) on ducklings. [Method] Ducklings of negative control group were fed with saline orally while ducklings in treatment group were infected with PM orally and instilled intranasally. The incidence and clinical symptoms of ducklings were observed. The ducklings in both groups were killed randomly at several times after infection. The livers,lungs,hearts,spleens,and thymus were collected by conventional paraffin section and H. E. staining and their histopathological changes were observed. The distribution of the PM in bodies of ducklings was studied with IFA (indirect fluorescent antibody test). [Result] Symptoms such as doldrums, dyspnea,and diarrhea were observed after PM was inoculated. Pathological changes included hydropericardium,lung bleeding and liver pitting necrosis. The dynamic variation about PM reproduction of were in thymuses for both methods. Histopathological changes were characterized by hyperemia and hem orrhage in the lung, fatty degeneration in the liver,and splenic cord abnormality. IFA positive signals main ly distributed in the lung,liver, thymus, and spleen. [Conclusion] Changes of hemorrhage and sepsis were found on ducklings infected by PM. Lung,liver, thymus and spleen were the main target organs.
