摘 要:【目的】研究鹿茸对小鼠脑组织抗氧化指标和凋亡相关蛋白表达的影响,为鹿茸的抗氧化和抗凋亡作用研究提供资料。【方法】选用48只6周龄雌性昆明小鼠,随机分为正常组(灌胃生理盐水,n=12)和高、中、低剂量鹿茸冻干粉组(分别灌胃鹿茸冻干粉1,3和5g/kg,n=12)。各组分别灌胃63d,第64天处死小鼠并取材。用试剂盒检测小鼠脑组织匀浆液中单胺氧化酶(Monoamine oxidase,MAO)、超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)的活性和丙二醛(Malondialdehyde,MDA)的含量,并采用免疫组化观察小鼠大脑皮层促凋亡蛋白Bax和抗凋亡蛋白Bcl-2的表达情况。【结果】鹿茸冻干粉高剂量组MAO活性极显著低于对照组和中、低剂量组(P<0.01),鹿茸冻干粉中剂量组MAO活性极显著低于对照组和低剂量组(P<0.01);鹿茸冻干粉高剂量组GSH-Px活性极显著高于对照组和中、低剂量组(P<0.01);鹿茸冻干粉高剂量组Bcl-2蛋白表达量极显著高于对照组和中、低剂量组(P<0.01),鹿茸冻干粉低剂量组Bcl-2与Bax表达量的比值显著低于对照组和高、中剂量组(P<0.05)。【结论】高剂量(5g/kg)鹿茸能提高机体抗氧化酶SOD和GSH-Px活性,降低MAO活性和脂质过氧化物MDA含量,增加抗凋亡蛋白Bcl-2的表达,具有增强机体抵抗生化损伤、清除体内脂质过氧化物、抗凋亡的作用。[Objective] This study focused on effects of Pilose antler on antioxidation indexes and pro- tein expression of apoptosis and anti-apoptosis in mice brain. [Method] 48 female Kunming mice with age of 6 weeks were randomly divided into 4 groups, control group (intragastric injection of saline, n= 12), lowdosage,medium-dosage and high-dosage groups of freeze-drying P. antler (intragastric injection of freeze-drying P. antler 1,3 and 5 g/kg,respectively,n=12 for each group). After being intragastric administrated for 63 days, the mice were killed for measurement of activity of monoamine oxidase (MAO), superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) and content of malondialdehyde (MDA) on morning of 64^th day. Immunohistochemical observation of the cerebral cortex was used to measure the expression of protein Bax and Bcl-2. [Result] The content of MAO in high-dosage group of freeze-drying P. antler was significant lower than that of other groups (P〈0.01). The higher the dosage,the lower the MAO content. The activity of GSH-Px in high-dosage group of freeze-drying P. antler was significant higher than other groups (P〈0.01). Protein expression of Bcl-2 in high-dosage group of freeze-drying P. antler was significant higher than other groups (P〈0.01). The protein expression ratio of Bcl-2 and Bax in low-dosage group of freeze-drying P. antler was significant lower than other groups (P〈0.05). [Conclusion] Statistics showed that high-dosage (5 g/kg) of P. antler can inhibit biochemical damage by promoting activity of SOD and GSH-Px,decreasing activity of MAO,reducing the content of internal lipid peroxide MDA,and increasing anti-apoptotic protein expression of Bcl-2.
