摘  要:采用PCR/DGGE技术和16SrDNA序列分析方法,对2种猪(普通猪、藏猪)肠道细菌优势菌群结构进行了比较分析。研究结果表明,从DGGE图谱中均可以发现2种猪肠道细菌具有一定的相似性,种内个体间相似性明显高于种间相似性。同时进行了部分优势细菌16SrDNA基因V6~V8区序列的系统发育分析发现,DGGE图谱中优势条带的16SrDNA基因序列中有7条克隆的序列与基因库最相似菌的相似性大于97%,其余的克隆序列相似性在90%~96%,其中有1条序列被鉴定为与纤维素分解菌产琥珀酸厌氧螺菌相似。本试验分析结果可为进一步研究藏猪及其肠道微生物的特性及其食草机理提供依据。DNA extraction from the rumen of two species of pigs (common pig and Tibet pig) was followed by polymerase chain reaction (PCR) amplification of 16S rDNA genes. PCR products were compared by denaturing gradient gel electrophoresis(DGGE) to compare the predominant bacterial community structure. The results showed the bacteria community structure of different goats was similar to each other. The similarities within species were noticeably higher than that between species. Phylogenetic and sequence similarity analyses of the resultant 29 clone sequences in 16S rDNA DGGE libraries revealed that 7 clone show similarity over 97% to that of database sequences,while the rest present similarity in a range of 90%-96%, and 1 clone was similar to Anaerobiospirillum succiniciproducens. The results lay the foundation for development of the Ti- bet big and its intestinal microflora.
