雌二醇在有序介孔碳修饰玻碳电极上的电化学行为及其测定

摘　　要：目的:采用硬模板法合成有序介孔碳(ordered mesoporous carbon,OMC),研究雌二醇(estradiol,E2)在有序介孔碳修饰玻碳电极(OMC/GCE)上的电化学行为,建立定量测定E2含量的电化学分析新方法。方法:循环伏安法(cyclic voltammetry,CV)和方波伏安法(square wave voltammetry,SWV)。结果:采用CV法,在0.1 mol·L-1的磷酸盐缓冲溶液(pH 8.0)中,E2在OMC/GCE上于+0.53 V处出现一不可逆的氧化峰,其氧化峰电流是GCE上的10倍。用SWV法测得E2氧化峰电流与其浓度在2.0×10-7～2.5×10-5mol·L-1呈线性关系,检出限(S/N=3)为1.0×10-7mol·L-1。采用本方法对女性血清中的E2进行加标回收率测定,回收率在97%～104%之间。结论:OMC/GCE对E2的电化学氧化具有良好的电催化作用,该方法可用于女性血清中E2的加标回收率测定。Objective：To synthesize ordered mesoporous carbon （OMC）by hard template method, study electro- chemical behaviors of estradiol （ E2 ） at the ordered mesoporous carbon modified glassy carbon electrode （ OMC/ GCE）, and establish a novel method for the determination of E2. Methods： Cyclic voltammetry （ CV ） and square wave voltammetry（SWV） were adopted. Results：The CV curves of E2 at OMC/GCE in 0. 1 mol · L-1phosphate buffer solutions （ PBS, pH 8.0） presented an irreversible oxidation peak at ＋ 0. 53 V, and the oxidation peak current of E2 at OMC/GCE was 10 times of that at GCE. Linear responses of E2 was obtained in the concentration range of 2.0 × 10^-7 -2. 5 ×10^-5 mol· L-lwith a detection limit of 1.0 × 10^-7 mol ·L-1. The proposed modified electrode was applied to the determination of E2 in female blood serums by standard addition method with the recovery in the range of 97% -104%. Conclusion： OMC/GCE exhibits strong electrocatalytic activity toward the oxidation of E2, and can be applied to the determination of E2 in female blood serums with satisfying results.
