摘  要:目的研究细胞周期蛋白依赖性激酶10在瘢痕疙瘩中的异常表达及其在瘢痕疙瘩发生机制中的作用。方法应用反转录聚合酶链式反应(1it.PCR)及Western Blot检测细胞周期蛋白依赖性激酶10mRNA及细胞周期蛋白依赖陛激酶10蛋白在人正常皮肤成纤维细胞及人瘢痕疙瘩成纤维细胞中的表达;将外源重组性真核质粒pCMV6-细胞周期蛋白依赖性激酶10转染到人瘢痕疙瘩成纤维细胞内为实验组;转染空质粒细胞及未处理细胞为对照组。用MTF法检测瘢痕疙瘩成纤维细胞增殖状况。结果与正常皮肤成纤维细胞相比,细胞周期蛋白依赖陛激酶10mRNA和细胞周期蛋白依赖f生激酶10蛋白在瘢痕疙瘩成纤维细胞中的表达显著下调,差异具有统计学意义(P〈0.01);与空pCMV6组和未转染组相比,pCMV6-细胞周期蛋白依赖性激酶10组细胞生长明显受到抑制,差异具有统计学意义(P〈0.01)。结论CDKl0在瘢痕疙瘩中呈下调表达,以CDKl0为靶点的基因治疗有潜在的临床应用前景。Objective To investigate the abnormal expression of cyclin-dependent kinase 10 in Keloid and the roles in mechanism of the keloid production pathogeny. Methods The expression of cyclin-dependent kinase 10 mRNA and protein in normal skin fibrobtast and Keloid fibroblast was measured by reverse transcription- poly- merase chain reaction (RT-PCR) and Western Blot, respectively. A recombinant plasmid carrying the cyclin-de- pendent kinase 10 gene (pCMV6-cyclin-dependent kinase 10) was transfected into keloid fibroblast as the experi- mental group, and keloid fibroblast untransfected and those transfected with empty pCMV6 plasmid were used as the control group. MTI" was applied to measure the effect of cyclin-dependent kinase 10 on the proliferation of Ke- loid fibroblasts. Results Compared with normal skin fibroblast, the expression of cyclin-dependent kinase 10 mRNA and protein was down-regulated in Keloid fibroblast ( P 〈 0.01 ). The growth of keloid fibmblast in the pCMV6-cyclin-dependent kinase 10 group was significantly lower than that in the empty pCMV6 and untransfeeted groups( P 〈 0.01 ). Conclusion cyclin-dependent kinase 10 is down-regulated in Keloid fibroblast compared with normal skin fibroblast. The CDK10 gene is a potential target for gene therapy of Keloid.
