摘 要:目的探讨原核基因重组脂联素球状结构域(gAd)对T2DM大鼠血糖水平的影响。方法采用高糖高脂饮食加1%STZ 35mg/kg建立T2DM大鼠模型。将造模成功的T2DM大鼠随机分为4组,检测干预前后血糖水平和40min后肝、肌肉组织己糖激酶(HK)和丙酮酸激酶(PK)活力。结果gAd干预40min时,干预组2、3血糖低于对照(NC)组,干预40min后,大鼠肝、肌肉组织HK和PK活力升高,差异有统计学意义(P<0.05)。结论 gAd急性干预T2DM大鼠可通过增强肝、肌肉组织糖酵解途径降低血糖水平。Objective To investigate the effect and its mechanism of globular domain of adiponectin (gAd) on the plasma glucose level in T2DM rat. Methods Rat models of T2DM were established by sucrose and fat-enriched diet and intraperitoneal administration of low-dose streptozotocin (STZ, 35 mg/kg). The T2DM rats were randomized into four groups. Plasma glucose levels in 0, 10, 20, and 40 min after intervention were measured, and the HK and PK vitality in the liver and muscle tissue were detected 40 rain after intervention. Results At the time of 40 min after intervention, the plasma glucose level in the gad intervention group 2 and 3 was lower than that in the NC group. At the time of 40 min after intervention, the increase of HK and PK vitality in the liver and muscle tissue of T2DM rats was statistically significant (P〈0.05). Conclusion Administration of gAd to T2DM rats reduces the plasma glucose level by enhancing the glycolysis in liver and muscle tissue.
