橡胶草胶乳蛋白双向电泳方法的建立

摘　　要：采用Tris饱和酚法和TCA/丙酮沉淀法提取橡胶草(Taraxacum Kok-saghyz Rodin)胶乳蛋白质,并建立蛋白双向电泳方法。结果表明,TCA/丙酮沉淀法得到的双向电泳图谱横条纹较多,背景模糊,得到约446个蛋白质点;Tris饱和酚法提取样品双向电泳的聚焦效果较好,图谱显示约630个蛋白点,其中包括较丰富的低分子量蛋白。The total protein samples of Taraxacum Kok-saghyz Rodin （TKS） latex extracted by Tris-saturated phenol method and trichloroacetic acid （TCA）/acetone precipitation method were analyzed by two-dimensional electrophoresis （2-DE）. The 2-DE gel of TCA/acetone precipitation showed strong horizontal streaks and background blur, and 446 protein spots; While the Tris-saturated phenol extraction was capable of generating more than 630 protein spots, obtaining better effect of isoelectric focusing. There were more small proteins revealed by Tris-saturated phenol extraction.
