应用空斑减少中和试验法对我国流行性出血热病毒进行血清学分型

摘　　要：应用空斑减少中和试验对分离自不同动物宿主和病人血液的流行性出血热病毒进行病毒抗原性分型,以不同株的家兔免疫血清对国际上血清1型和2型参考毒株进行试验。结果表明,3株来自黑线姬鼠的毒株,其免疫血清对血清1型(76-118株)的效价较血清 2型(UR株)高≥16倍,而6株来自家鼠和大白鼠的毒株中,有5株对2型的血清效价高于1型2～16倍。提示来自黑线姬鼠的毒株符合国际上的血清1型,来自褐家鼠的毒株基本与国际上的血清2型一致,但个别毒株例外。以上结果表明,空斑减少中和试验特异性高,可用于流行性出血热病毒的准确血清学分型。 本文还发现,J10株(分离自锦州市褐家鼠)和HB55诛(分离自河南出血热病人)的血清型与过去文献报道的不符。经反复用不同免疫血清和型特异性单克隆抗体做空斑减少中和试验,均表明这两株病毒的分型与血清1型相同。以上结果为我国首次应用空斑减少试验对我国出血热不同毒株进行准确分型的报道。Serotypic classification of HFRS viruses isolated from rodents and patients were investigated using plaque reduction neutralization test ( PRNT ) . Sera of rabbits inoculated with different HFRS viruses were tested against reference type-1 Hantaan virus and type-2 Seoul virus. The neutralizing titers of antisera of Apod emus isolates against type-1 Hantaan virus were 16-fold higher than those for type-2 Seoul virus, while the antisera of Rattus isolates had titers to type-2 viruses 2-16-fold higher than those for type-1 viruses. So different isolates could be classified into serotype 1 (consistent with Hantaan virus) and serotype 2 (consistent with Seoul virus) .We also found two isolates, one from Rattus, another from patient, which could be classified into serotype 1 by PRNT, which is inconsistent with data previously published.
