鸡新城疫病毒单克隆抗体的中和作用

摘　　要：23株抗鸡新城疫病毒(NDV)单克隆抗体(简称单抗)中,9株(10-1、15-1、L15-6、LA15-7、FHY-23、39-S-2、44-S-2、94-St 107-2具有中和能力。研究证明,有溶血抑制能力的单抗均有中和能力,表明病毒的溶血能力与感染性相关。该9株单抗对吸附前的NDV F48E8株有不同程度的中和作用,蚀斑减少中和试验、病变抑制中和试验和鸡胚中和试验的中和效价呈平行相关性。单抗L15-6对已吸附于细胞的F48E8 4℃作用1小时,使感染细胞荧光灶形成单位(ICFFU)减少了77.6％。单抗10-1、15-1、L15-6、LA15-7等分别作用于感染了F48E8的细胞,能抑制此感染性细胞产生病变.免疫荧光试验证实,在病变受抑制的细胞内仍有病毒抗原,一旦终止单抗的连续作用,孔内细胞在48小时内即产生病变。单抗对NDV的中和作用,可能包括阻止病毒的吸附、穿入或促使已吸附的病毒解离等,但单抗并不能完全中和细胞内存在的病毒。单抗15-1、L15-6均能保护SPF鸡不受100LD50 F48E8的攻击,15-1还对已出现鸡新城疫症状的实验性病鸡有治疗作用。A panel of 23 hybridoma cell lines were used to prepare McAb specific for Newcastle disease virus ( XDV ). Nine of them showed neutralizing activity. All the McAbs inhibiting hemolysis had neutralizing capability, indicating that virus infectivity is correlated with the ability of virus to induce cell fusion. Incubation of NDV with neutralizing McAbs resulted in various degree of neutralization. Neutralizing titers in plaque redaction neutralization, cytopathic effect ( CpE ) inhibition and chick embryo neutralization assays were found to be parallel. The infectious cell fluorescent focus formation units ( ICFFU ) was reduced up to 77.6% when virus adsorbed cells were incubated with L15-6 for lhr at 4℃. CPE was inhibited by 4 McAbs(10-1, 15-1, L15-6 and LA15-7) when they were added to NDV-infected Vero cells. Immunofluorescent assay showed that virus antigen still existed in the cytoplasm of CpE inhibited cells, and CpE re-emerged in 40hr after replacement with medium without McAbs. It was postulated that McAbs could neutralize NDV by inhibiting virus adsorbtion and penetration as well as by releasing adsorbed virus from cells. Virus present in the cells could not be completely neutralized by McAbs. McAbs 15-1 and L15-6 were capable of protecting Sp F chicks aganist F48-Es virulent strain challenge at a dose of 100 LD50. McAb 15-1 showed promising therapeutic effect ia chicks even after clinical signs of experi-mantal Newcastle disease ( ND ) had appeared.
