摘 要:目的 :探讨 pSVPoMcat微基因修饰雪旺氏细胞 (SC)对损伤脊髓再生修复的作用。方法 :采用切割法制备健康SD大鼠脊髓半横断损伤模型 ,随机植入 pSVPoMcat微基因修饰的SC(A组 ) ,高纯化的SC(B组 ) ,仅植入明胶海绵的损伤对照组 (C组 )。受体鼠存活 3月 ,行MRI扫描 ,取材作电镜 (TEM )观察。结果 :MRI发现 :A组动物 ,脊髓损伤 (SCI)区脊髓信号已基本恢复正常 ,B组动物未恢复正常 ,而C组动物 ,SCI区有软化灶形成。TEM发现 :A组动物SCI区大量有髓和无髓轴突再生 ,SC增殖 ;B组动物SCI区 ,可见有髓轴突出再生部分SC已变性坏死 ;C组动物 ,SCI区未见有髓及无髓轴突。结论 :pSVPoMcat微基因修饰SC移植对SCI有促进修复的作用 。Objective: To approach the effect of genetically modified Schwann cell (SC) with pSVPoMcat in the regeneration of spinal cord injury (SCI). Methods: Experimental animal was divided into three groups: the pSVPoMcat genetically modified SC implantation group (A group), SC implantation group (B group), and control group (C group). After three month the presence of axonal regeneration of the injuried spinal cord was examined by means of MRI and TEM. Results: The results were found: (1) The MRI spinal cord signal of SCI region has basic recovered in A group and has not recoved completely in B group. There was a malacia focus in C group. (2) The outcome of TBM observation in A group, B group was purification SC and regenerated axons, necrotic SC partially and regenerated axons, respectively. Otherwise, there were not regenerated axons in C group. Conclusions: This result is considered to promote axonal regeneration and potential for further clinical application.
