流行性出血热(EHF)地鼠肾细胞(GHKC)灭活疫苗的初步人体观察

摘　　要：用流行性出血热(EHF)病毒L99株(家鼠型)感染金黄地鼠肾细胞(GHKC),经福尔马林(0.025％)灭活制备EHF灭活疫苗(88-17批),经检定批准,肌肉(1ml)接种志愿者12人(IO人于0、7、28天,2人分别于0、7天及0、28天各一针),进行了安全性及抗体反应的初步人体观察。结果显示,此种疫苗对人体接种无明显不良反应。接种后,12名接种者IFA、ELI-SA及中和抗体(EFRNT)全部阳转,180天和360天后仍分别有10人血清中和抗体阳性。初步表明,此疫苗是安全的,可以有效诱导抗体产生,其中和抗体多数可持续一年以上。The inactivated vaccine against epidemic hemorrhagic fever ( EHF ), Lot 88-17,was prepared with golden hamster kidney cell ( GHKC) culture, infected with the rat-type EHF virus, L99 strain, and inactivated with 0.025% formalin. 12 human volunteers were immunized with the vaccine, 10 of them on 0, 7, 28 days, and 2 on 0,7 days,and 0 , 28 days respec- tively, in order to assess the safety and ,the antibody responses of this vaccine. The results showed that, no obvious untoward reaction was observed. The vaccinees were followed up by successive bleedings after 28, 42, 180 and 360 days. The total possitive sero-conversion rates of IFA, ELISA antibodies were 100%, and that of neutralizing antibody ( EFRNT ) was also 100%, including sera of the two vaccinees received two doses of the vaccine. There were sera of 10 out of these 12 vaccinees, collected both on 180 days and 360 days later were still positive for neutralizing antibody. These data demonstrated that the vaccine was safe and could effectively induce specific antibody responses.
