摘 要:目的探讨荆芥、桂枝挥发油含药血清体外抗甲型流感病毒(H1N1)的作用。方法采用细胞病变法(CPE)检测荆芥、桂枝挥发油含药血清对狗肾传代细胞(MDCK)的最大无毒浓度(TC0);采用血凝试验测定甲型流感病毒对MDCK细胞的感染性;采用血凝试验和四甲基偶氮唑蓝(MTT)比色法测定荆芥、桂枝挥发油含药血清对甲型流感病毒的体外抑制作用。结果①荆芥、桂枝挥发油含药血清对MDCK细胞的TC0均为5%,即在相应培养基中血清含量为5%。②甲型流感病毒鼠肺适应株A/PR/8/34(H1N1)对MDCK细胞的TCID50(病毒半数组织感染量)为10-3.5/0.1 ml。③荆芥、桂枝挥发油含药血清5%、3.5%、2.5%浓度对10 TCID50的甲型流感病毒的增殖均有显著抑制作用,且5%、3.5%浓度的荆芥、桂枝挥发油含药血清对甲型流感病毒具有直接杀灭作用。结论荆芥、桂枝挥发油含药血清对甲型流感病毒在MDCK细胞中的增殖具有显著抑制作用,并具有一定直接杀灭作用,结果与挥发油的体外、体内实验一致,进一步明确证实了两者的抗流感病毒效应。Abstract : Objective To study the anti - influenza A virus ( IAV, H1 N1 ) effect of the serum containing volatile oil of Schizonep- etae and Ramulus Cinnamomi in vitro. Methods The TC0 of the serum containing volatile oil of Schizonepetae and Ramulus Cinna- momi in MDCK ceils was determined by CPE assay. The infectivity of IAV to MDCK cells was determined by hemagglutination test. The IAV inhibitory effect of the serum in vitro was determined by hemagglutination test and MTY colorimetric assay. Results The TC0 of the serum containing volatile oil of Schizonepetae and Ramulus Cinnamomi in MDCK cells both were 5%, which means the concentration of serum in reaction system should be less than 5%. The TCIDs0 of mouse - lung adapted strain of IAV A/PR/ 8/34( I-I1N1 ) to MDCK cells was 10-35/0. lml. 5%, 3.5%, 2.5% serum containing volatile oil of Schizonepetae and Ramulus Cinnamomi significantly inhibited the multiplication of IAV in MDCK cells, while the 5% , 3.5% serum also showed the direct killing action to IAV. Conclusion The serum containing volatile oil of Schizonepetae and Ramulus Cinnamomi could significantly inhibit IAV multiplication in MDCK cells, while showed the direct killing action to IVA. The results were similar to the effect of valotile oil in vitro and in vivo, which confirmed the anti - ilffluenza virus action of two drugs.
