摘  要:目的探讨斑蝥素盐类衍生物——斑蝥素酸镁在体外对肺癌细胞增殖的抑制作用。方法采用磺酰罗丹明染色法(SRB法)考察斑蝥素酸镁对H-1299和A-549两种人肺腺癌细胞的体外抗肿瘤活性;采用流式细胞术测定斑蝥素酸镁对H-1299细胞周期的影响;利用细胞集落形成实验,检测斑蝥素酸镁对单个肿瘤细胞形成克隆的影响。结果斑蝥素酸镁对H-1299、A-549两株肿瘤细胞具有比较明显的抑制效果,其半数抑制浓度(IC50)分别为1.59μg.ml-1、1.39μg.ml-1,且抑制率随药物浓度的增加而升高,呈剂量效应关系。细胞周期实验显示H-1299细胞在受试药物的作用下S期和G2/M期的细胞比例呈明显减少(P<0.05),G0/G1期细胞比例显著增加(P<0.05),细胞周期阻滞于G0/G1期。细胞集落形成实验结果表明,当斑蝥素酸镁的浓度达到2.10μg.ml-1时,肿瘤克隆就不能形成。结论斑蝥素酸镁对肺腺癌细胞具有非常明显的抗肿瘤活性,可做进一步研究。Objective The objective of this study was to evaluate the anti-tumor effects of magnesium cantharidate on human lung adenocarcinoma cell lines in vitro. Methods The sulforhodamine B(SRB) assay was employed to evaluate the anti-tumor effects of magnesium cantharidate on H-1299 and A-549 cell in vitro.Flow cytometry was used to measure the induction of cell cycle arrest.Colony forming assay was conducted to detect the cloning influence of magnesium cantharidate on individual tumor cells. Results The results showed that magnesium cantharidate has obvious inhibitory effects on the proliferation of H-1299 and A549 cell in a dose-dependent manner.The IC50 value of H-1299 and A-549 were 1.59 μg·ml-1 and 1.39 μg·ml-1 respectively.The cell cycle of H-1299 treated by magnesium cantharidate changed.The ratio of cultured cell declined in S and G2/M phases(P<0.05),while the cell in G0/G1 phase increased dramatically.The cell cycle arrested in G0/G1 phase.Colony forming assay showed that the cloning of the single H-1299 cell could not happen if the magnesium cantharidate's concentration was to achieve 2.10 μg·ml-1. Conclusion Magnesium cantharidate has strong anti-tumor effects in vitro,which is worth probing further.
