摘  要:目的研究Smad3在通脉大生片作用后的大鼠卵巢颗粒细胞中的表达。方法将培养的原代大鼠卵巢颗粒细胞爬片后,随机分为自然生长对照组、通脉大生片高剂量组、通脉大生片中剂量组、通脉大生片低剂量组、FSH组,分别在细胞生长对数期给药,取给药后的细胞爬片,用免疫组化法检测细胞爬片中Smad3的表达量。结果自然生长对照组各时间点的Smad3的表达量无明显变化,通高组、通中组、通低组给药12h时Smad3核阳性率明显高于给药6h和24h(P<0.05);通中组给药12h时Smad3核阳性率明显高于通高组和通低组12h时Smad3核阳性率,有统计学意义(P<0.05);FSH组给药后1h、2h时Smad3核阳性率明显高于正常组,有统计学意义(P<0.05),FSH组给药后2h时Smad3核阳性率明显高于给药后1h时Smad3核阳性率,有统计学意义(P<0.05)。结论通脉大生片可能是通过增加Smad3的表达量来促进卵泡发育,以达到治疗排卵障碍性不孕的目的 。Objective To study the expression of Smad3 in ovarian granulosa cells. Methods ovarian granulosa cells in slides were cultured and were randomly divided into control group, high - dose group, medium - dose group, low - dose group and FSH group, respectively to dose the medicine inthecytoplasmie growth logarithmic phase, detect the expression quantity of Smad3 by immunohistochemistry. Results the expression of Smad3 at each time point did not change significantly in the natural growth con- trol group, the nuclear positivity of Smad3 after administration of 12h in high -dose group, medium -dose group and low - dose group was significantly higher than that of after administration of 6h and 24h ( P 〈 0.05) ; the positive rate of Smad3 after admin- istration of 12h in medium - dose group was significantly higher than that nuclear positivity of Smad3 after administration of 12h in high nuclear group and through the low group, with statistical significance ( P 〈 0.05 ) ; the nuclear positivity of Smad3 after ad- ministration of 1 h,2h inFSH group was significantly higher than that of normal group, there were statistically significant (P 〈 0.05 ), the nuclear positivity of Smad3 after administration of 2h in FSH group was significantly higher than that after the adminis- tration of 1 h, with statistical significance (P 〈 0.05 ). Conclusion TMDST may be increased expression quantity of Smad3 to promote the follicular development, in order to achieve the purpose of treating anovulatory infertility.
